摘要
目的 建立血浆中罗格列酮及其代谢产物的定量测定方法 ,为药物代谢研究提供技术手段。方法 采用高效液相色谱方法 ,ZorbaxODS(5 μm ,15 0× 4 .6mm)色谱柱 ,甲醇 - 0 .0 5mol·L-1乙酸钠溶液 (pH6 .0 ) (4 6∶5 4 )为流动相 ,紫外检测波长为2 4 7nm ,血浆样品经碱化乙酸乙酯提取 ,以内标法峰面积定量。结果 检测下限 0 .0 5 μg·ml-1,方法线性范围为 0 .0 5~ 10 .0 0 μg·μml-1;在 0 .1、0 .5、2 .5、5 .0 μg·ml-1四种浓度平均回收率为 80 .4 5 % ;日内、日间平均RSD分别为 0 .98%、2 .2 8%。结论 该方法灵敏度高 ,专一性强 ,可满足药代动力学研究的要求。
Aim To develop a HPLC assay method for the determination of rosiglitazone and its metabolite in dog plasma.Methods Zorbax ODS colurm(5μm,150×4.6mm)as the stationary phase and methanol-0.05mol·L -1 sodium acetate(pH6.0)(46∶54) as mobil phase,the UV determination wavelength of 247nm.The plasma sample was extrscted by ethyl acetate.Results The minimum detection concentration was 0.05μg·ml -1 .The method average extraction recovery was 80.45%.The inter-day and intra-day RSD was less than 4.6%.Conclusion The assay method was sensitive,accuracy and applicable for pharmacokinetic study.
