金银花在体内外抗人Ⅰ型疱疹病毒的实验研究

Experimental Studies on Flas Lonicerae Anti-Herpesvirus I in Vivo and in Vitro

目的 :为了观察金银花在体内外的抗人疱疹病毒Ⅰ型作用 ,我们进行了此项研究。方法 :我们以阿昔洛韦 (ACV)为阳性对照 ,以PBS为阴性对照 ,用HSV 1HS 1株感染豚鼠皮肤的动物模型 ,以及HSV 1F株和HSV 1HS 1株感染的Hep 2细胞为实验系统 ,用空斑减数实验 ,观察了金银花抗HSV 1药效。结果 :在体外实验中 ,ACV对HSV 1F株和HSV 1HS 1株增殖抑制的ED5 0分别为 9 0 9μg ml ,2 0 0 4 μg ml。金银花对HSV 1F株直接杀灭、感染阻断、增殖抑制的ED5 0依次为8 36 μg ml、3 2 2 μg ml、3 76 μg ml;对HSV 1HS 1株的相应数值分别为 10 84 μg ml、10 0 4 μg ml、6 35 μg ml。结果 :动物实验临床观察表明 ,ACV、金银花处理皮区的累积计分依次为 14 2 0± 6 70 ;7 4 6± 1 75 ;ACV、金银花处理皮区的痊愈天数分别为 7 70± 1 0 8;7 30± 1 2 0 ,与PBS处理皮区有明显差异 ,并具有统计学意义。HSV 1HS 1株感染过程中各处理皮区的大体照片、皮肤样本的病毒滴度得到了一致的结果。结论 :体外实验的结果表明 ,金银花对HSV 1F株和HSV 1HS 1株增殖抑制的治疗指数分别是ACV相应指标的 10 4 ,72倍 ,药效比ACV好。配对T检验的结果表明 ,ACV与金银花处理皮区的累积计分、痊愈天数无显著差异。病毒滴度检测结果、大体照片?

Objective:In order to investigate antiherpes effects on human simplex virus type 1 of Lonicera japonica Thunb in vitro and in vivo,we carried out this research.Methods:We used Acyclovir as positive control,and used PBS as negative control,used guniea pigs model of cutaneous herpes virus infection,and Hep 2 cells infected by HSV 1F and HS 1 strain as experimental systems,and used plaque reduction assays,observed the antiherpes effects of Lonicera japonica Thunb.Results:In in vitro experiments,the ED50s of the inhibition effects on the multiplecation of HSV 1F and HS 1 strain of ACV were 9.09 μg/ml,20 04 μg/ml respectively.The ED50s of direct virucidal activity,the inhibiton effects on the multiplication and infection of HSV 1F strain of Lonicera japonica Thunb were 8 36μg/ml,3 22μg/ml,3 76μg/ml separately,and the corresponding figures to HSV 1HS 1strain of it were 10 84μg/ml,10 04μg/ml,6 35μg/ml respectively.The results of animal experiments showed that the cumulative scores dealt with ACV,Lonicera japonica Thunb were 14 20±6 70;7 47±1 75 respectively,and the time to healing days of the regions dealt with ACV,Lonicera japonica Thunb were 7 70±1 08;7 30±1 20 separately,these results had significances in statistics with the regions dealt with PBS.The pictures of each treatment regions and the measurements of virus titres of skin samples got same results.Conclusions:The in vitro experiments showed that the Treatment Indices of inhibition effects on the multiplication of HSV 1F and HS 1 strain of Lonicera japonica Thunb were 104 and 72times of same parametres of ACV,it's drug effects was better than ACV.The results of Student's T test suggested that the cumulative scores and time to healing days of skin regions dealt with ACV had no differences in statistics with the cumulative scores and time to healing days of skin regions dealt with Lonicera japonica Thunb,these results added up the results of measurements of virus titres,the pictures of guniea pigs infected with HSV 1HS 1 strain,to suggest the antiherpes effects of the Lonicera japonica Thunb were close to ACV,and should be worth exploitation.
