摘要
目的建立犬骨质疏松实验动物模型并初步观察纳米级羟基磷灰石(HA)涂层人工关节置换后的效果。方法选择8只平均年龄5.5±0.38岁、平均体重为16±2.5kg的雌性犬,分成实验组和对照组,实验组切除双侧卵巢,与对照组在同一环境下饲养24w。手术切除犬右侧股骨头,然后分别装入普通柄人工关节或纳米级HA涂层人工关节。12w后分别取每只犬的双侧股骨(一侧含人工关节)、胫腓骨、尺桡骨以及以椎骨进行骨密度检测,X光双侧股骨摄片检查及血液生化检查。结果两组动物模型骨标本骨密度检测以及X光双侧股骨摄片显示,去卵巢组犬符合重度骨质疏松症,而人工关节周围骨密度均匀不等,其中普通柄人工关节周围骨组织结构紊乱,含纳米级HA涂层人工关节柄周围结构良好。结论采用切除双侧卵巢的方法成功地建立了犬骨质疏松模型并初步发现纳米级HA涂层人工关节较普通柄人工关节的置换效果好。
Objective To construct an osteoporosis dog model and preliminary teat the application of nanometer HA on artificial joint replacement. Methods Eight female dogs of average age 5.5±0.38 years old and average weight 16±2.5 kg were divided into control group and experimental group. The experimental group was ovariotomized and kept in the same condition with the control group for 24 weeks. The right femur was removed and replaced with ordinary artificial join or HA coated artificial join. After operation the dogs were not limited to move and were feed for twelve weeks in the same condition. Dogs were then killed and the two femurs(one with artificial joint), shin fibula, radius and lumbar vertebrae were obtained for the measurement of bone density, X ray check up and histological studies. Results The bone mineral density (BMD) examination and X ray examination of both side of femurs showed that dogs with ovariotomy resembled of those with serious osteoporosis. The bone density around the artificial join was not even, especially in the ordinary artificial join. The structure of the nanometer HA coated artificial join was smooth and in good condition. Conclusion Demonstrated that ovariotomized dog is a good osteoporosis model and nanometer HA coated artificial join is better than those with common handle.
