摘要
目的 检测细胞凋亡相关基因TNFAIP3和ENC1在不同级别胶质瘤组织和正常脑组织中的表达。方法 提取组织总RNA,用半定量RT-PCR检测其中TNFAIP3和ENC1 mRNA的表达。结果 (1)同一患者肿瘤组织与其正常脑组织相比,TN-FAIP3基因表达上调,ENC1基因表达下调;(2)与正常脑组织相比,WHO Ⅱ和Ⅳ级肿瘤组TNFAIP3表达升高,WHO Ⅰ、Ⅱ和Ⅲ级肿瘤组ENC1表达下降。结论 RT-PCR结果与芯片检测结果基本相符;胶质瘤组织中TNFAIP3表达上调、ENC1表达下调,它们的异常表达可能参与了胶质瘤的恶性进展过程。
Objective To detect the expression of apoptosis-related genes TNFAIP3 and ENC1 in different grade glioma specimens and normal brain tissues. Methods Extracting total RNA of tissues, then detecting the expression of TNFAIP3 and ENC1 with semi-quantitative RT-PCR. Results (1) TNFAIP3 was up-regulated and ENC1 down-regulated in glioma tissue comparing with normal brain tissue of same individual. (2) Compared with normal brain tissues, TN-FAIP3 was increased in WHO Ⅱ and Ⅳ groups, and ENC1 was decreased in WHO Ⅰ, Ⅱ, and Ⅲ groups. Conclusions The results of gene expression in RT-PCR were similar with cDNA array. The abnormal expression of TNFAEP3 and ENC1 may be involved in malignant progression of glioma.
