摘要
目的 :探讨可溶性支架转染c myc反义寡核苷酸是否可抑制静脉移植物内膜的增殖。方法 :2 5只健康纯种新西兰大耳白兔按体重随机分为对照组 ,支架组 ,正义组 ,反义组 ,错配组 ,将颈外静脉间置于同侧颈总动脉后用可溶性支架将反义 ,正义及错配的c myc基因片段导入静脉移植物。术后 2 8d取静脉桥标本 ,测定内膜、中膜厚度。结果 :反义组静脉桥的内膜厚度为 (4 3.77± 5 .89) μm ,显著低于对照组、支架组、正义组及错配组 ((80 .0 0±1.34) μm ,(76 .0 6± 4 .2 4 ) μm ,(74 .80± 9.6 4 ) μm ,(79.6 2± 7.92 ) μm ,P <0 .0 1) ;反义组内膜与中膜比值为0 .6 8± 0 .2 0 ,明显低于对照组、支架组、正义组及错配组 (1.4 2± 0 .18,1.35± 0 .2 0 ,1.34± 0 .2 6 ,1.4 1± 0 .2 4 ,P <0 .0 1)。结论 :运用可溶性支架转染反义c
Aim: To evaluate whether intimal hyperplasia can be inhibited through transcatheter delivery of c myc ODNs with soluble stents. Methods: Twenty five New Zealand rabbits were randomized into five groups(five in each group):control group,stent group,sense group,antisense group, and mismatch group. External jugular vein were interposed between ipsilateral common carotid arteries. Sense group,antisense group, and mismatch oligonucleotides were locally administered through soluble stents.Animals were killed at the 28th days after operation and defined segments of vein grafts were sectioned and observed under microscope.The intimal,medial thickness and ratios of intimal to medial thickness were measured with the aid of computer. Results:The intimal thickness in antisense groups ((43.77±5.89) μm) significantly decreased compared with those in control,stent,sense, and mismatch groups ((80.00±1.34) μm,(76.06±4.24) μm,(74.80±9.64) μm ,(79.62±7.92) μm, respectively ( P <0.01)), and there was no significant difference among these 4 groups.The ratios of intimal to medial thickness for antisense groups (0.68±0.20)significantly decreased compared with those in control,stent,sense, and mismatch groups (1.42±0.18, 1.35±0.20, 1.34±0.26, 1.41±0.24,respectively, P <0.01.Conclusion: Intimal hyperplasia of rabbit vein graft can be markedly suppressed by intravascular local delivery of synthetic antisense c myc via soluble stent.
