摘要
目的 研究铜绿假单胞菌对亚胺培南耐药的遗传学机制。方法 建立检测铜绿假单胞菌编码IMP型金属酶基因和外膜蛋白OprD_2基因的PCR法,探讨临床分离的16株纸片协同法产金属酶的铜绿假单胞菌中产金属酶和外膜蛋白OprD_2的缺失与亚胺培南耐药的关系。结果 16株产金属酶铜绿假单胞菌IMP基因扩增7株呈阳性,9株阴性;OprD_2基因扩增11株呈阴性,5株呈阳性;11株菌既产金属酶,又存在外膜蛋白OprD_2基因突变。结论 产金属酶和外膜蛋白OprD_2的缺失是铜绿假单胞菌对亚胺培南耐药的重要机制,并可同时并存。
Objective To study the genetic mechanism of imipenem-resistance in Psedomonas aeruginosa. Methods PCR methods was established to detect the IMP gene of metallo-p-lactamases and outer mem-brance protein D2 (OprD2 ) gene in Psedomonas aeruginosa. The relationship between the detection of metallo-p-lactamases and the loss of OprD2 and imipenem-resistance were investigated in 16 strains of clinically isolated metallo-p-lactamases-producing Psedomonas aeruginosa. Results seven of 16 strains of metallo-B-lactamas-es-producing Psedomonas aeruginosa were positive for IMP gene; OprD2 gene was negative in 11 strains and positive in 5 strains. Both metallo-B-lactamases production and the loss of OprD2 were found in 11 strains. Conclusion Production of metallo-B-lactamases and the loss of OprD2 are the essentianl mechanisms of imipenem-resistance in Psedomonas aeruginosa and can coexist.
