摘要
以4个番茄品种为材料,通过对子叶和茎段培养,建立了番茄组织培养的再生体系,为番茄的遗传转化提供了良好的受体系统;通过农杆菌介导,初步将HIV-1的gag基因、gp120基因和gag-gp120嵌合基因导入番茄,得到了抗性转化再生植株,再生植株PCR检测结果呈阳性,从而建立了良好的番茄遗传转化体系。
The leaves and stem segments of four tomato cultivars as explants were cultured. The results indicated that the leaves and stem segments induced callus. The callus further differentiated buds. After the buds produced shoots, plantlets were regenerated. Tomato culture system was established. Tomato was transformed via Agrobacterium tumefaciens (EHA105 and LBA4404) harboring plasmids carrying gag, gp120 and gag-gp 120 genes of HIV-1. Transformants were obtained in the experiment. It was demonstrated that gene gag had been inserted into tomato genome after PCR assay. Factors influencing tomato transformation were studied. Genetic transformation system of tomato mediated by Agrobacterium tumefaciens was built. These could afford the basic research for transgenic tomato as bioreactor to produce oral vaccines.
