摘要
从 4 0个PstI/MseI酶切的AFLP引物对中筛选出多态性高的 7个 ,对 36份枳属特异种质进行了指纹分析。 7个引物对共获得 5 39条带 ,其中 14 6条带具有多态性。各种质材料之间的遗传相似性在 0 2 3~ 0 98之间 ,只需结合其中的 3个引物对即可区分所有的材料。根据 7个AFLP引物对所得谱带的UPGMA聚类分析表明 ,聚类结果与地理来源没有明显的联系 ,这可能正反映了近时间内地区之间枳资源的频繁交流。
AFLP(Amplified Fragment Length Polymorphism)analysis system was established on Poncirus accessions.Seven primer pairs with high polymorphism have been screened from 40 primer pairs.The screened subset of primer pairs produced 539 bands,146 of which were polymorphic.Genetic similarity among 36 accessions ranged from 0 23 to 0 78.All the 36 accessions could be clearly differentiated based on the bands obtained from three of the seven primer pairs.This provided a first example to distinguish all tested Poncirus accessions. The genetic distance was calculated and a dendrogram was constructed based on AFLP data of 7 primer pairs.The separation of Poncirus accessions did not correspond to their geographical distribution,which may result from the frequent exchange of seeds of Poncirus plants among different regions recently.
