非小细胞肺癌中血管内皮标记物显示微血管密度的对比研究

Comparison of CD_(34) , CD_(31) and F_(VIII) served as labels to show the microvascular density in non-small cell lung cancer

目的 探讨血管内皮标记物CD3 1、CD3 4 和FVIII在显示非小细胞肺癌 (NSCLC)微血管密度(MVD)中的差异 ,以及MVD与淋巴结转移的关系。方法 采用微波免疫组化染色方法 (LSAB)检测CD3 1、CD3 4 与FVIII在 78例NSCLS中的表达。结果 血管内皮标记物CD3 4 、CD3 1与FVIII显示的MVD分别是 (6 5 8± 14 3)、(6 3 5± 17 9)和 (5 4 3± 19 4 )条 ,范围分别是 18～ 112条、16～ 10 2条和 16～ 10 3条 ,具有一致性 (P >0 0 5 )。CD3 4 、CD3 1与FVIII3种微血管内皮标记物显示NSCLC的MVD ,在转移组分别是(6 9 5± 15 7)、(6 5 3± 14 5 )和 (5 8 3± 16 3)条 ,在无转移组分别是 (4 4 3± 14 3)、(4 0 5± 12 6 )和 (36 5± 15 8)条 ,转移组与无转移组间差异均有非常显著性 (P <0 0 1)。结论 CD3 1、CD3 4 、FVIII相关抗原均能清晰地、选择性地显示NSCLC的MVD ,但CD3 4 比CD3 1、FVIII因子相关抗原更敏感 ,能突出显示较小的、不成熟的微血管或单一的内皮细胞 ;NSCLC的MVD与微血管内皮标记物的种类无关 ,与有无转移密切相关 ,MVD可作为预测NSCLC病人预后的指标。

Objective: In order to evaluate the difference among CD 34 , CD 31 and F VIII in showing the microvascular density(MVD) in non small cell lung cancer (NSCLC) and the relation of MVD to metastasis of tumors. Methods: The LSAB immunohistochemical technique was used to investigate the expression of CD 34 , CD 31 and FVIII in 78 cases of NSCLC. Results: The MVD in NSCLC shown by CD 34 , CD 31 and VIII related antigen labeling were 65 8±14 3 (ranged from 18 to 112), 63 5±17 9 (ranged from 16 to 102) and 54 3±19 5 (ranged from 16 to 103) respectively with a similar tendency (P>0 05). CD 34 , CD 31 and F VIII of the average counts of microvessels was 69 5±15 7, 65 3±14 5 and 58 3±16 3 in the NSCLC with metastasis and 44 3±14 3, 40 5±12 6 and 36 5±15 8 in those without metastasis showing a significant difference (P<0 05). Conclusion: The endothelial cell of microvessels could be shown clearly by its related antigen labeling such as CD 34 , CD 31 and F VIII . CD 34 was more sensitive than CD 31 and F VIII . It could show smaller, immatured microvessels, and even single endothelial cell. The MVD was related closely to lymph node metastasis (P<0 01). It could be used as one of the indexes for predicating the prognosis.
