摘要
目的 探讨非洛地平对 5 / 6肾大部分切除大鼠心肌细胞L型钙电流的影响。方法 5 / 6肾大部分切除手术制作肾功能衰竭大鼠模型 ,使用膜片钳全细胞记录技术记录心肌细胞L型钙电流。结果5 / 6肾切除大鼠心肌细胞L型钙电流明显增大 ,稳态失活曲线左移。非洛地平能够逆转L型钙电流的增大 ,但未能逆转通道失活性质的改变。结论 慢性肾衰竭可能引起心肌细胞钙操纵的改变 ,非洛地平可以逆转这种改变 ,还能促进L型钙离子通道的电压依赖性失活。
Objective To investigate the effects of felodipine on L-type calcium currents of rat cardiomyocytes after 5/6 subtotal nephrectomised. Method Animals were divided into three groups as follow: gorup A, 5/6 subtotal nephrectomy(SNx);group B, SNx administrated with 10mg/kg/d of felodipine;gorup C,control.Sprague-dawley rats underwent two-stage SNx; control rats underwent bilateral renal decapsulation. Animals were sacrificed after 12 weeks.Whole-cell patch clamp techniques were used to examine L-type calcium currents(ICa,L)in isolated cardiomyocytes. Results The amplitude of L-type calcium current significantly increased in SNx rats and was blocked by felodipine partly.The potential giving 50%of inactivation(V1/2) moved towards more negative in SNx rats than in control,and it had no change after administrated felodipine.Conclusions The calciumhandling of SNx rats cardiomyocytes changes.Felodipine can reverse this change of calcium handling partly and promote the voltage-dependent inactivation of L-type calcium channel.
