摘要
目的 :探讨牛膝多糖免疫调节作用的机制。方法 :使用牛膝多糖在不同浓度或在不同时间内对人T细胞进行体外诱导培养 ,用ELISA方法测定培养上清液的IFN γ及IL 4的蛋白表达情况。结果 :①人T细胞受不同浓度ABPS刺激时 ,IFN γ分泌水平随ABPS浓度递增 ,在 4 0 0 μg ml时 ,IFN γ表达量最高 (P <0 0 5 ) ,而IL 4的分泌始终处于低水平 (P >0 0 5 )。②人T细胞在ABPS刺激不同时间后 ,IFN γ分泌水平逐步增高 ,在 4 8小时时与其他各时间点比较 ,有非常显著的差异 (P <0 0 0 1) ,而IL 4的分泌始终处于低水平 (P >0 0 5 )。结论 :①ABPS能诱导人T细胞分泌IFN γ ,该作用呈时间、剂量依赖性变化 ,而抑制IL 4的分泌。②在蛋白表达水平上证实ABPS能够促进Th1类细胞因子的分泌 ,而抑制Th2类细胞因子的分泌。
Objective:To study the immunity modulation mechanism of Achyranthes bidentata polysaccharides.Methods:The T cells freshly isolated in 24 well flat bottom plates cells were cultured with serially diluted ABPS for 48 h or with 400 μg/ml ABPS for various time at 37℃ in 5% CO 2.Supernatants were assay for IFN γ and IL 4 using ELISA kit.Results:①IFN γ protein levels were continued to increased together with the increasing concentration of ABPS. IFN γ protein levels were most when the concentration of ABPS is 400 μg/ml(P<0.05). But IL 4 protein levels were always low compared with that of control group(P>0.05). ②T cells were cultured with ABPS for 8,18,24 h and 48 h, IFN γ protein was continued to increase, IFN γ protein levels at 24~48 h elevated fast ,and at 48 h significantly compared with that of other time(P<0.001).But IL 4 protein levels were also always low compared with that of control group(P>0.05).Conclusion:①ABPS can induce secretion IFN γ protein, and the effect is time and dose dependent, and ABPS can anti induce the expression of IL 4 protein. ②ABPS may up regulate Th1 type cytokine expression and down regulate Th2 type cytokine expression at protein level. [
