葛根芩连汤不同配伍情况下葛根素、黄芩苷、小檗碱的含量比较 被引量:24

摘要 目的 :比较葛根芩连汤不同配伍情况下葛根素、黄芩苷、小檗碱的含量变化。方法 :采用反相高效液相色谱法。选用HypersilC18柱 ,流动相分别以甲醇 -水 (30∶70 ) ,在2 5 0nm波长测定葛根素 ;甲醇 - 0 0 4 %磷酸 (4 6∶5 4) ,在 2 80nm波长测定黄芩苷 ;乙腈 -0 0 2mol/L磷酸二氢钾 (35∶6 5 ) ,在 346nm波长测定盐酸小檗碱。结果 :3种成分的峰都能有效分离。线性范围分别为葛根素 0 0 74 1~ 2 2 2 μg ,黄芩苷 0 0 2 80~ 4 6 7μg ,盐酸小檗碱0 15 7~ 3 13μg。结论 :中药复方煎煮过程中存在动态变化 ,应根据不同的目的对样品进行适宜的处理。 Objective: To establish a method for the identification of the content change of Kakonein, Baicalin and Berberine Hydrochloride in different compatibility by HPLC. Methods: The included chromatographic conditions were Hypersil C18 column and mobile phases consisted of a mixture of methanol-water (30:70) for kakonein, methanol- 0.04% phosphoric acid (46:54) for baicalin and acetonitrile-0.02mon/L KH2PO4 (35:65) for berberine hydrochloride. The detection wavelengths were at 250nm, 280nm and 346nm respectively. Results: A good linearity was found in the concentration range of 0.0741~2.22μg for kakonein, 0.0280~4.67μg for baicalin and 0.157~3.13μg for berberine hydrochloride respectively. The correlation coefficients were 1.0000, 1.0000 and 0.9998, respectively. The average recovery of kakonein was 96.30% with RSD 0.59%, that of baicalin was 101.16% with RSD 1.20%, and that of berberine hydrochloride was 99.84% with RSD 2.75%. Conclusion: The method was found to be simple and accurate. The contents of different constituents in different compatibility were different.
