摘要
以双子叶植物中的十字花科芸苔属油菜(Bassica Campestros L.)为试验材料,探讨了水合法、冷冻酶解法、直接酶解法、水合-酶解二步法和三步法等多种破壁方法,比较得出油菜成熟花粉,水合法破外壁最为理想,而其中以1mol/L蔗糖为水合液最好,其破外壁率可达92.4%,油菜幼嫩花粉的破壁以水合-酶解二步法最好,其中以0.8mol/L甘露醇+2%纤维素酶组合最好,其破壁率达85.7%。
Manifold ways of breaking the cell wall were discussed in the paper, using Brassica Campestros L. of dicotyledon as the material. We compared the different projects of breaking the outer cell wall and the inner cell wall of the mature pollens and the young pollens of the cole. This test provided the optimum technical basis for the using of nutriment and the protoplast of the pollen.
