苹果汁中展青霉素和羟甲基糠醛的高效液相色谱测定方法 被引量:17

High Performance Liquid Chromatography Method for Determining Patulin and HMF in Apple Juice

摘要 为了建立能有效地将苹果汁中羟甲基糠醛分离的展青霉素高效液相色谱测定方法,将苹果汁样品用乙酸乙脂提取,提取液经碳酸钠溶液净化,无水硫酸钠脱水,再通过反相色谱柱检测。色谱柱为supelco C18柱,紫外检测器检测波长为275nm。结果表明:展青霉素和羟甲基糠醛能完全分离,其检出限均为5μg/L,加标回收率分别为73.1%~82.7%和78.8%~83.6%。从而建立了一个能有效分离羟甲基糠醛的准确、灵敏的展青霉素高效液相色谱测定方法。 In order to establish a method for determining patulin in apple juice by high performance liquid chromatography (HPLC) which can separate 5-hydroxymethyl-2-furaldehyde (HMF) effectively, the apple juice sample was extracted with ethyl acetate and the extract was cleaned up with sodium carbonate solution and then was dehydrated by anhydrous sodium sulfate. Patulin and HMF were determined by reversed-phase HPLC using a supelco C18 column and an ultraviolet detector at 275nm. The results showed that Patulin and HMF could be completely separated and the detected limits for patulin and HMF were equal 5μg/L. The recoveries for patulin and HMF were 73.1%-82.7% and 78.8%-83.6% respectively. In the present study, an accurate, sensitive method for determining patulin by HPLC, which can separate HMF effectively, has been established.
