摘要
研究了咕吨类染料丁基罗丹明B(BRB)与DNA作用的共振光散射光谱.加入DNA后,在pH1.5~2.5的范围内,BRB在DNA分子表面发生长距离自组装,在400nm处产生了增强的共振光散射峰,其发光强度与DNA的浓度成线性关系,线性范围为5.8~2688ng·mL-1,检出限(3σ)为5.8ng·mL-1.考察了影响因素和最佳反应条件,建立了用共振光散射谱RLS测定纳克级DNA的新方法.
Resonance light-scattering (RLS) spectra of butyl rodamin B (BRB) with DNA was studied. When DNA was added, long range assembly of BRB on the molecular surfaces of nucleic acid occurred in the pH1.5~2.5, resulted in an enhanced resonance light-scattering peak at 400?nm. The intensity of resonance light-scattering was found to be proportional to the concentration of DNA in the linear range 5.8~2?688 ng·mL-1, and the determination limit (3σ) is 5.8 ng·mL-1.The effects of interference substances and optimization of procedure were investigated and a new method for determination of the DNA at nanogram level based on RLS measurement was established.
