沙利度胺对人宫颈癌细胞增殖抑制的研究

摘要 目的:探讨沙立度胺对人宫颈癌Hela细胞增殖抑制的作用。方法:体外培养宫颈癌Hela细胞株,加入不同浓度的沙利度胺(50、100、200ug/ml),同时设空白对照组;采用MTT法测量各组细胞增殖抑制率,Western-blot法检测细胞中环氧化酶-2(COX-2)的变化。结果:沙利度胺对人宫颈癌Hela细胞的生长具有明显的抑制作用,并随药物浓度和时间的增加实验组中细胞的抑制率逐渐增加,较对照组差异有显著性(P<0.05);实验组中COX的表达均低于对照组(P<0.05)。结论:沙利度胺对宫颈癌Hela细胞的生长具有抑制作用,其机制可能与下调COX-2有关。 Objective: To investigate the effect of thalidomide on proliferation of human cervical cancer Hela cells. Methods: The cervical cancer cells line Hela cultured in vitro, with different concentrations of thalidomide(50, 100, 200 ug/ml), and the blank control group; cells proliferation inhibition rate was measured by MTT method, Western-blot assay of cyclooxygenase-2(COX-2) changes. Results: Thalidomide can obviously inhibit the growth of human cervical carcinoma Hela cells, and increase the drug concentration and time of suppressor cells in the experimental group increased, compared with the control group the difference was significant(P<0.05); the expression of COX in experimental group was lower than the control group(P< 0.05). Conclusion: The growth of cervical cancer Hela cells was inhibited by thalidomide, and the mechanism may be related to down-regulation of COX-2.
