姜黄素干扰瘢痕疙瘩成纤维细胞内TGF-β_1/Smad信号转导的研究 被引量:1

摘要 目的:研究姜黄素对瘢痕疙瘩成纤维细胞(keloid fibroblasts,KF)TGF-β_1/Smad信号转导途径的影响方法:免疫沉淀,western blot检测姜黄素对KF内Smad2/3磷酸化,Smad2/3/4复合物表达的影响。结果:姜黄素明显抑制TGF-β_1诱导的pSmad2L, pSmad3L的表达,而对pSmad2C, pSmad3C表达的抑制作用相对较弱,对pSmad2L和pSmad3C的抑制作用具有浓度依赖性。姜黄素呈浓度依赖性显著抑制Smad2/3/4复合物表达,在15,30mg/L浓度上几乎完全阻断复合物形成。结论:姜黄素可能通过干扰TGF-β_1/Smad信号转导途径,发挥抗瘢痕疙瘩作用。 AIMS To investigate the effects of curcumin on TGF-β1/Smad signal transduction pathway in KF.Methods Smad2/3 phosphorylation and Smad2/3/4 complex formation in KF were detected by immunoprecipitation and western blot.Results Curcumin significantly inhibited TGF-β1-induced pSmad2 L and pSmad3 L expression, whereas exhibited relative weaker effects on pSmad2 C and pSmad3 C. Inaddition, the expression of pSmad2 L and pSmad3 C,the Smad2/3/4 complex expression induced by TGF-β1 was markedly supressed by curcumin in a dose-dependent manner, and almost entirely blocked at the concentration of 15 or 30 mg/L.Conclusions curcumin might exert anti-keloid effects via interfering with TGF-β1/Smad signal transduction pathway.
