嗅鞘细胞移植治疗帕金森病动物模型的相关研究

摘要 目的利用大鼠同种异体嗅鞘细胞移植的方式对帕金森病模型大鼠进行治疗,初步探究在PD模型大鼠脑内移植嗅鞘细胞能否成为治疗帕金森病的一种有效手段。方法提取和培养原代OECs,利用6-OHDA两点注射法建立帕金森病动物模型,通过APO诱导方式验证模型及检测治疗效果,通过HE染色进行大鼠黑质细胞形态学观察,利用抗TH荧光染色细胞计数对比治疗前后TH阳性神经元表达情况差异。结果从嗅鞘细胞移植后的第4周开始,PD模型的行为学诱导旋转圈数明显减少。HE染色结果示:PD模型大鼠毁损侧的黑质致密部的神经元变性明显,且TH阳性神经元数量降低,经OECs治疗后的帕金森大鼠的患侧黑质部位神经元变性改善,且TH阳性神经元含量明显增高。结论嗅鞘细胞能够改善帕金森模型大鼠的行为学变化。嗅鞘细胞能够提高帕金森模型大鼠患侧黑质部位多巴胺神经元含量。 Objective Olfactory ensheathing cellhomotransplantations were performed on Parkinson’s disease rat models,to preliminarily explore whether olfactory ensheathing cell transplantation into the brain of PD rat models can be an effective method for the treatment of Parkinson’s disease. Methods We extracted and cultivated original generation of OECs,and established animal model of Parkinson’s disease by injecting 6-OHDA at two points,then We validated the model and detected the therapeutic effect by APO induced rotation method,observed the morphology of rat nigral cells by HE staining,and compared the expression of TH positive neurons before and after treatment by anti-TH fluorescence staining cell count. Results From the fourth week after olfactory ensheathing cell transplantation,the number of ethologically induced rotations in PD model decreased significantly. HE staining results showed that the neurons in the dense part of the damaged substantia nigra denatured obviously in PD model rats,and the number of TH positive neurons decreased. After OECs treatment,the denaturation of neurons in the substantia nigra of PD rats was improved,and the content of TH positive neurons significantly increased. Conclusion Olfactory ensheathing cells can improve the behavioral changes of Parkinson’s disease model rats. Olfactory ensheathing cells can increase the dopamine neurons content in the substantia nigra at the affected side of Parkinson’s disease model rats.
