摘要
目的:观察加味益气聪明汤对血管性痴呆(VD)大鼠行为学及海马区胆碱能神经递质乙酰胆碱(Ach)、胆碱酯酶(Ach E)的影响。方法:60只实验大鼠随机分为假手术组、模型组、尼莫地平组(简称"对照组")、加味益气聪明汤组(简称"实验组")。采用一次性永久性结扎双侧颈总动脉(2-VO)法制备VD模型;尼莫地平组予以6.25mg/kg/d,加味益气聪明汤组予以14.58g/kg/d,均按所需剂量配制成2ml/d的浓度进行给药,模型组、假手术组予以2ml/d蒸溜水灌胃,一共持续给药30天,1次/天。选用Morris水迷宫实验检测大鼠的行为学变化,用ELISA法测定海马区Ach、Ach E的含量。结果:与假手术组比较,模型组大鼠逃避潜伏期(EL)明显延长(P<0.01),经过有效区及平台的次数显著减少,停留在平台及有效区的时间显著缩短(P<0.01),海马区Ach含量明显降低,海马区Ach E含量明显增多(P<0.01)。与模型组比较,对照组、实验组EL明显缩短(P<0.05或P<0.01),经过平台及有效区的次数显著增多,平台及有效区停留时间明显延长(P<0.05或P<0.01);对照组与实验组比较,经过平台及有效区的时间以及跨越平台及有效区的次数无明显变化,差异无统计学意义(P>0.05)。与模型组比较,对照组、实验组海马区Ach含量显著增多,而Ach E含量明显降低,差异有统计学意义(P<0.01)。与对照组比较,实验组大鼠海马区Ach含量略有降低,Ach E含量略有增高,但差异无统计学意义(P>0.05)。结论:加味益气聪明汤能改善VD大鼠的学习记忆功能,其机制与提高大鼠海马区Ach含量,降低Ach E含量有关。
Objective: Investigating the influence of Modified Yiqi Congming Decoction on behavior of VD rats and observing the changes of hippocampus neurotransmitter acetylcholine( Ach) and cholinesterase( AchE) content in rats. Methods: Dividing randomly the 60 male SD rats into sham operation group,model group,Nimodipine group( controlled group),Modified Yiqi Congming Decoction group( experimental group). The operation group with an operation had left and right carotid artery ligation( CCA) for preparing for VD model( CCA). The rats in the Nimodipine group were given Nimodipine in the dose of 6. 25mg/kg/d; the rats in the experimental group were given Modified Yiqicongming Decoction in the dose of 14. 58 g / kg / d; all drugs were made up 2ml / d according to each requirement dosage. The rats in the sham operation group and model group were given aqua water at 2ml / d,lasting for 30 days,one time for one day. Detecting each group behavior mainly through the water maze experiment;detecting the content of hippocampus Ach and AchE were detected by the means of the ELISA method. Results: To compare with the sham operation group,the rats in the model group whose escaping latency( EL) were prolonged( P < 0. 01),the platform and effective regional time shortened obviously,and the number of striding across the platform and effective area were significantly reduced( P <0. 01); to compare with the model group,the rats in the controlled group and the experimental group,whose EL were obviously shortened( P <0. 05 or P <0. 01),crossing the number of platform and effective area were increased significantly and the time of staying on the platform and the effective residence were significantly prolonged( P <0. 05 or P < 0. 01),and the content of Ach were increased markedly but the content of AchE were decreased markedly( P <0. 01). There was no great difference on the time of staying on the platform and the number of striding across the platform and effective area between the controlled group and the experimental group,it was not statistical meaning( P > 0. 05). To compare with the model group,the content of Ach in hippocampus in the controlled group and the experimental group were increased markedly,but the content of AchE were decreased markedly in the meantime,there were statistical meaning( P <0. 01). To compare with the controlled group,the content of hippocampus Ach in the experimental group were decreased slightly while the content of hippocampus AchE were increased a little,but it was not statistical meaning( P >0. 05). Conclusion: Modified Yiqicongming Decoction can improve the function of learning and memory,and improve the content of the hippocampus Ach,but decrease the content of the hippocampus AchE.
