加味四妙散汤对急性痛风性关节炎大鼠炎性细胞因子及多靶点调节信号通路影响 被引量:16

Effect of Modified Simiao Powder Decoction on Inflammatory Cytokines and Multiple Target Regulatory Signaling Pathway for Rats with Acute Gouty Arthritis

摘要 目的:探讨加味四妙散汤对急性痛风性关节炎大鼠炎性细胞因子及多靶点调节信号通路影响。方法:选取由本院实验动物中心提供雄性Wistar健康大鼠40只,依据随机数字表法分为四组,对照组、模型组、秋水仙碱组与加味四妙散汤组,每组10只;制备急性痛风性关节炎大鼠模型,使用ELISA法检测各组大鼠关节液内炎症因子含量,免疫印迹法检测关节周边组织内Caspase-1、TLR4、ASC及NF-κB蛋白表达情况,RT-PCR检测关节周边组织内Caspase-1、TLR4、ASC及NLRP3mRNA表达情况。结果:模型组IL-6、TNF-α及IL-1β含量较对照组上升,加味四妙散汤组和秋水仙碱组较模型组下降,差异均有统计学意义(P<0.05),加味四妙散汤组TNF-α含量较秋水仙碱组升高,差异有统计学意义(P<0.05);模型组Caspase-1、TLR4、ASC及NLRP3mRNA表达量较对照组上升,加味四妙散汤组Caspase-1、ASC mRNA表达量较模型组下降,秋水仙碱组Caspase-1、TLR4、ASC及NLRP3mRNA表达量较模型组和加味四妙散汤组下降,差异均有统计学意义(P<0.05)。结论:加味四妙散汤可抑制急性痛风性关节炎大鼠关节液内炎性因子含量,同时调节信号通路内蛋白及mRNA表达水平起到治疗作用。 Objective: To analyze the effect of modified Simiao Powder Decoction on inflammatory cytokines and multiple target regulatory signaling pathway in rats with acute gouty arthritis. Methods: By the experimental animal center of our hospital to provide healthy male Wistar 40 rats,according to randomly divided into four groups,controlled group,model group,colchicine group and Modified Simiao Powder Decoction group,10 rats in each group; The preparation of model rats with acute gouty arthritis,using ELISA method to detect inflammation factor in synovial fluid of rats,the expression of Caspase-1,TLR4,ASC and NFB protein was detected by Western blot in the tissues surrounding the joint,the expression of Caspase-1,TLR4,ASC and NLRP3 mRNA to detect RT-PCR in the tissues surrounding the joint. Results: In the model group,IL-6,TNF-alpha and IL-1 beta content increased more than the controlled group,with the Simiao Powder Decoction group and the colchicine group decreased more than the model group,the differences were statistically significant(P<0. 05),the TNF-alpha content of the Modified Simiao Powder Decoction group increased more than the colchicine group,the difference was statistically significant(P<0. 05). The expression of Caspase-1 in the model group; TLR4,ASC,NLRP3 and mRNA increased compared with the controlled group,the expression of Caspase-1,ASC mRNA of the Modified Simiao Powder Decoction group decreased compared with the model group,the expression of Caspase-1,TLR4,ASC and NLRP3 mRNA of the colchicine group decreased more than the model group and the Modified Simiao Powder group,the differences were statistically significant(P<0. 05). Conclusion: Modified Simiao Powder Decoction can inhibit the inflammatory factors in the joint fluid of rats with acute gouty arthritis,and regulate the expression of protein and mRNA in the signaling pathway.
