摘要
目的:探讨丹栀逍遥散对多囊卵巢综合征模型大鼠的干预作用。方法:使用脱氢表雄酮(DHEA)诱导建立多囊卵巢大鼠高雄激素血症模型,并设立丹栀逍遥组、达英组、模型组、空白对照组,予以相应药物干预。检测大鼠体重变化、卵巢颗粒细胞卵泡刺激素受体(FSHR)、黄体生成素受体(LHR)蛋白表达情况和大鼠血清睾酮(T)、孕酮(P)、雌二醇(E2)、卵泡刺激素(FSH)及黄体生成素(LH)、水平变化。使用HE染色,观察大鼠卵巢细胞形态结构。结果:与空白组对比,模型组大鼠体重显著增加,且差异有统计学意义(P<0.05),与模型组相比较丹栀逍遥组大鼠的体重则显著降低,(P<0.05),且恢复到正常水平。大鼠体内FSHR含量显著升高,且差异有统计学意义(P<0.05),LHR含量显著升高,且差异有统计学意义(P<0.05),T含量显著降低,且差异有统计学意义(P<0.05),E2含量显著升高,且差异有统计学意义(P<0.05),P、FSH、LH含量则无显著变化(P>0.05)。与模型组相比,丹栀逍遥组大鼠、达英~35组大鼠卵泡膜细胞较少,无明显细胞凋亡的形态特征。结论:丹栀逍遥散通过促进大鼠细胞内FSHR、FSH蛋白的表达来降低大鼠体内雄性激素水平对多囊卵巢综合征具有调节改善作用。
Objective:To explore the intervention effect of Danzhi Xiaoyao Powder for model rats with polycystic ovary syndrome.Methods:Dehydroepiandrosterone(DHEA)was used to establish a model of hyperandrogenism in polycystic ovary rats.Danzhi Xiaoyao group,Diane group,model group and blank controlled group were established and given corresponding drug intervention.The body weight changes,protein expression levels of ovarian granulosa cell follicle-stimulating hormone receptor(FSHR)and luteinizing hormone receptor(LHR)and levels of serum testosterone(T),progesterone(P),estradiol(E2),follicle stimulating hormone(FSH)and luteinizing hormone(LH)were measured.The morphology of rat ovarian cells was observed by HE staining.Results:Compared with the blank group,the body weight in the model group was significantly increased(P<0.05),and the body weight in the Danzhi Xiaoyao group was significantly decreased compared with that in the model group(P<0.05),and returned to normal level.The FSHR level in rats was significantly increased(P<0.05),and the LHR level was significantly increased(P<0.05),and the T level was significantly decreased(P<0.05)while the E2 level was increased significantly(P<0.05),and there were no significant changes in the levels of P,FSH and LH(P>0.05).Compared with the model group,the quantity of theca cells was lower in the Danzhi Xiaoyao group and the Diane-35 group,and there were no obvious morphological features of apoptosis.Conclusion:Danzhi Xiaoyao Powder can reduce the regulation and improvement effect of androgens on polycystic ovary syndrome by promoting the protein expressions of FSHR and FSH in rats.
