宣肺活血汤对内毒素性肺损伤大鼠模型肺功能的保护作用

Protective Effect of Xuanfei Huoxue Decoction on Lung Function for Model Rats with Endotoxin-Induced Lung Injury

目的:探究宣肺活血汤对内毒素性肺损伤大鼠模型肺功能的保护作用及其机制。方法:将30只SD大鼠随机分为对照组、模型组和宣肺活血汤组(7.25mg/kg),各10只,采用尾静脉注射脂多糖(LPS)法构建内毒素性肺损伤大鼠模型。采用HE染色观察大鼠肺组织病理变化;观察大鼠肺泡灌洗液(BALF)中细胞总数和中性粒细胞数目;采用ELISA法检测大鼠超氧化物歧化酶(SOD)、丙二醛(MDA)、白介素-6(IL-6)及肿瘤坏死因子α(TNF-α)的含量;采用Western blot法检测大鼠肺组织中NF-κB、JNK、p38蛋白的表达。结果:与对照组相比,模型组大鼠BALF中细胞总数、中性粒细胞数目,血清中MDA、IL-6及TNF-α含量,肺组织中NF-κB、JNK、p38蛋白表达均显著上升,血清中SOD的含量显著下降(P<0.05);给予宣肺活血汤处理后,大鼠BALF中细胞总数、中性粒细胞数目,血清中MDA、IL-6及TNF-α含量,肺组织中NF-κB、JNK、p38蛋白表达均显著下降,血清中SOD的含量显著上升(P<0.05)。结论:宣肺活血汤可以减轻内毒素性肺损伤模型大鼠肺功能障碍,可能通过参与氧化应激和炎症反应途径改善肺损伤。

Objective:To explore the protective effect and mechanism of Xuanfei Huoxue Decoction on lung function in model rats with endotoxin-induced lung injury.Methods:30SD rats were randomly divided into controlled group,model group and Xuanfei Huoxue Decoction group(7.25 mg/kg),with 10 rats in each group.Endotoxin-induced lung injury rate model was established by injection of lipopolysaccharide(LPS)into the tail vein.The pathological changes of lung tissues were observed by HE staining.The total number of cells and the number of neutrophils in rat alveolar lavage fluid(BALF)were observed.The levels of superoxide dismutase(SOD)and malondialdehyde(MDA),interleukin-6(IL-6)and tumor necrosis factorα(TNF-α)were detected by ELISA.The expression levels of NF-κB,JNK and p38proteins in rat lung tissues were detected by Western blot.Results:Compared with the controlled group,the total number of cells and the number of neutrophils in BALF,the levels of serum MDA,IL-6 and TNF-αand the expression levels of NF-κB,JNK and p38proteins in lung tissues were significant increased while the level of serum SOD was decreased significantly(P<0.05)in the model group.After treatment with Xuanfei Huoxue Decoction,the total number of cells and the number of neutrophilsin BALF,the levels of serum MDA,IL-6 and TNF-αand the expression levels of NF-κB,JNK and p38proteins in lung tissues were decreased significantly while the level of serum SOD was increased significantly(P<0.05).Conclusions:Xuanfei Huoxue Decoction can alleviate lung dysfunction in rats with endotoxin-induced lung injury,and it may improve lung injury by participating in oxidative stress and inflammatory response pathways.
