清化固肾排毒方对腺嘌呤所致大鼠慢性肾功能衰竭的保护作用 被引量:3

摘要 目的:研究清化固肾排毒方对腺嘌呤所致大鼠慢性肾功能衰竭的保护作用。方法:选择SD大鼠60只,随机选取45只用0.5ml剂量250mg/(kg·d)的腺嘌呤连续灌胃21天构建大鼠慢性肾功能衰竭模型(CRF),15只为对照组灌胃等体积的蒸馏水,对比两组大鼠体重及生长状态。造模结束后各组随机选取5只大鼠,尾静脉取血分离血清检测大鼠血清中尿素氮(BUN)、肌酐(SCr)、尿酸(UA)及尿液中24小时尿蛋白水平。将模型大鼠随机分为15只/组随机分为模型组和清化固肾排毒方高、低剂量治疗组,分别接受2ml剂量为3g/kg、0.3g/kg的清化固肾排毒方颗粒灌胃治疗31天,对照组和模型组每日灌胃等体积蒸馏水。治疗结束后采集各组大鼠血清,检测BUN、Scr、UA及尿液中24小时尿蛋白水平;大鼠处死取肾组织观察病理变化,westernblot分析肾组织中TNF-a的表达量。结果:与对照组比较,模型大鼠生长构建期间生长状态明显变差,体重增长显著降低,血清中BUN、SCr、UV、24小时尿蛋白水平显著增高(P<0.05)。清化固肾排毒方治疗后,与模型组比较,大鼠血清中BUN、SCr、UV及尿液中24小时尿蛋白水平显著降低,且呈现出明显的剂量依赖性(P<0.05)。病理分解结果显示,治疗组随着剂量的增加肾组织的萎缩、扩张、炎性细胞浸润、纤维化症状明显改善,尿酸结晶沉淀明显减少,westernblot结果显示,与模型组比较,治疗组大鼠肾组织TNF-a、bFGF的表达水平呈现出显著的剂量依赖性降低(P<0.05)。结论:清化固肾排毒方能够有效治疗腺嘌呤所致大鼠慢性肾功能衰竭,其机制可能与降低肾脏组织TNF-a、bFGF的表达有关。 Objective:To study the protective effect of Qinghua Gushen Paidu Recipe for chronic renal failure rats induced by Adenine.Methods:60 SD rats were selected,and 45 rats of them were randomly selected and given continuous intragastric administration of 0.5 ml dose of 250 mg/(kg.d)adenine for 21 d to construct rats models of chronic renal failure(CRF),and another 15 rats were given intragastric administration of same volume of distilled water,and the weight and growth status of rats were compared between the two groups.After modeling,5 rats were randomly selected from each group,and their blood from tail vein was collected and serum was isolated,and the levels of blood urea nitrogen(BUN),creatinine(SCr)and uric acid(UA)in serum and 24 h urine protein in urine were measured.The models rats were randomly divided into model group and high and low dose Qinghua Gushen Paidu recipe group with 15 rats in each group,and treatment groups were given 2 ml doses of 3 g/kg and 0.3 g/kg of Qinghua Gushen Paidu Recipe intragastrically for 31 d,and the controlled group and the model group were intragastrically administrated with an equal volume of distilled water.At the end of treatment,the serum of each group was collected,and the levels of BUN,Scr and UA and 24 h urine protein in urine were detected.The rats were sacrificed and renal tissues were collected to observe the pathological changes,and the expression level of TNF-a in renal tissues was analyzed by Western blot.Results:Compared with the controlled group,the growth status of model rats was significantly worse during the growth and construction period,and the weight gain was significantly decreased,and the levels of serum BUN,SCr,UV and 24 h urine protein were significantly increased(P<0.05).After treatment with Qinghua Gushen Paidu Recipe,the levels of BUN,SCr and UV in serum and 24 h urine protein in urine were significantly decreased,and showed dose-dependence compared with the model group(P<0.05).The results of pathological decomposition showed that with the increase of dose in the treatment group,the atrophy,dilation,inflammatory cell infiltration and fibrosis of renal tissues were significantly improved while the uric acid crystal precipitation was significantly reduced.Western blot results showed that compared with the model group,the expression levels of TNF-a and bFGF in renal tissues in the treatment groups showed a significant dose-dependent decrease(P<0.05).Conclusion:Qinghua Gushen Paidu Recipe can effectively treat rats with chronic renal failure induced by adenine,and its mechanism may be related to the decrease of TNF-a and bFGF expression in renal tissues.
