乌司他丁对肠缺血再灌注损伤大鼠小肠细胞凋亡的影响 被引量:5

摘要 目的探讨乌司他丁对肠缺血再灌注损伤大鼠的保护作用及其机制。方法将60只健康Wistar大鼠随机分为对照组(A组)、肠缺血再灌注后0 h组(B1组)、肠缺血再灌注后2 h组(B2组)、乌司他丁治疗肠缺血再灌注后0 h组(C1)、乌司他丁治疗肠缺血再灌注后2 h组(C2组)各12只。B1、B2、C1、C2组夹闭肠系膜上动脉(SMA)60 min制作肠缺血再灌注损伤模型,并于手术前经尾静脉B1、B2组各注入生理盐水2 m L,C1、C2组各注入乌司他丁5×104U/kg;A组仅分离SMA,不夹闭血管。取A、B1、B2、C1、C2各组大鼠的小肠组织,光镜下观察组织学改变,TUNEL法检测小肠细胞凋亡率。结果光镜观察发现,A组小肠黏膜基本正常;B1组小肠黏膜及绒毛排列轻度紊乱,无黏膜变性、坏死,出现上皮下间隙扩大,腺体轻度受损及毛细血管充血;B2组小肠绒毛水肿明显,黏膜及绒毛排列明显紊乱,黏膜上皮脱落呈现糜烂,腺体严重受损;以上变化在绒毛顶端尤为显著。C1、C2组小肠黏膜充血、水肿及上皮糜烂坏死的程度均较B1、B2组减轻。B1、B2组大鼠细胞凋亡率较A组升高(P均<0.01)。C1组与B1组、C2组与B2组比较细胞凋亡率均降低(P均<0.05)。结论乌司他丁可抑制小肠细胞凋亡,有助于减轻由于缺血再灌注引起的肠道损伤。
