白藜芦醇对游离脂肪酸诱导的胰岛β细胞凋亡的影响 被引量:1

摘要 目的观察白藜芦醇(Res)对游离脂肪酸(FFA)诱导的胰岛β细胞凋亡的影响。方法将对数生长期的胰岛β细胞随机分为A^F组,A、B组分别置于不含FFA及含500μmol/L棕榈酸的培养基中培养,C^F组分别置于含0.1、1、10、50μmol/L的Res+棕榈酸500μmol/L的培养基中培养24 h,采用流式细胞术测算细胞存活率及细胞凋亡率。结果 A^F组细胞存活率分别为90.31%±4.70%、5.07%±2.27%、8.37%±2.06%、21.99%±2.47%、88.67%±1.17%、5.20%±1.72%,细胞凋亡率分别为7.40%±2.99%、95.82%±1.94%、83.36%±2.06%、68.94%±2.19%、11.03%±1.08%、95.65%±1.72%,B^F组与A组比较,P均<0.05;C^E组与B组比较,P均<0.01;D、E组与C组比较,P均<0.01;D、E组间比较,P均<0.01。结论 FFA能促进体外培养的β细胞凋亡,Res在一定浓度范围内能拮抗棕榈酸诱导的胰岛β细胞凋亡。 Objective To evaluate the effects of resveratrol( Res) on the apoptosis of β cells exposed to palmitic acid( PA,a free fatty acid). Methods β cells in the period of logarithmic phase were randomly divided into A-F groups. A group was blank group excluding PA,and B group was cultured with 500 μmol / L PA. C-F groups were put in culture medium with 0. 1,1,10,50 μmol / L Res + 500 μmol / L PA respectively. After 24 hours,flow cytometry was used to calculate the survival rate and the apoptosis rate of β cells. Results The survival rate of A-F groups were 90. 31% ± 4. 70%,5. 07% ± 2. 27%,8. 37% ± 2. 06%,21. 99% ± 2. 47%,88. 67% ± 1. 17%,5. 20% ± 1. 72%,while the apoptosis rates were 7. 40% ± 2. 99%,95. 82% ± 1. 94%,83. 36% ± 2. 06%,68. 94% ± 2. 19%,11. 03% ± 1. 08%,95. 65%± 1. 72%,respectively. There were significant differences among A group and B-F groups( all P < 0. 05). Compared with B group,C-E group displayed remarkably lower apoptosis rates and higher survival rates( all P < 0. 01). There were significant differences among C group and D,E groups( all P < 0. 01); and compared with D group,E group displayed lower apoptosis rate and higher survival rate( all P < 0. 01). Conclusion PA increases β cell apoptosis in vitro,Res at some appropriate concentrations may reduce β cell apoptosis induced by PA.
