褪黑素对PANC-1细胞增殖的影响及机制探讨

摘要 目的观察褪黑素对人胰腺癌细胞(PANC-1细胞)增殖的影响,并探讨其机制。方法取PANC-1细胞,以不同浓度褪黑素干预,采用MTT法检测PANC-1细胞增殖情况并计算增殖率,Western blotting法检测PANC-1细胞中Notch1的活性片段(NICD1)。结果对照1组细胞增殖率为0.38%±0.03%,褪黑素组加入1、1 000、1 000 000 nmol/L的褪黑素后细胞增殖率分别为0.36%±0.02%、0.32%±0.02%、0.26%±0.01%,对照1组与褪黑素组加入1 000、1 000 000 nmol/L的褪黑素后增殖率比较,P均<0.05。对照2组细胞增殖率为0.43%±0.02%,DAPT组细胞增殖率为0.30%±0.02%,两组比较,P<0.05。对照1组NICD1相对表达量为0.47±0.05,褪黑素组加入1、1 000、1 000 000 nmol/L的褪黑素后NICD1相对表达量分别为0.32±0.04、0.33±0.05、0.21±0.03,对照1组与褪黑素组加入1 000 000 nmol/L的褪黑素后NICD1相对表达量比较,P均<0.05。对照2组NICD1相对表达量为0.63±0.05,DAPT组NICD1相对表达量为0.38±0.03,两组比较,P<0.05。结论褪黑素可以抑制PANC-1细胞的增殖,其机制可能是通过调节Notch信号通路来实现。 Objective To observe the influence of melatonin on the PANC-1 cell proliferation and to investigate its mechanism. Methods PANC-1 cells were treated with different concentrations of melatonin,and MTT method was used to detect the proliferation of PANC-1 cells and to calculate the proliferation rate. Western blotting was used to detect the expression of active fragment of Notch 1( NICD1) in PANC-1 cells. Results The proliferation rate of the control group 1was 0. 38% ± 0. 03%,after adding the concentration of 1,1 000 and 1 000 000 nmol / L melatonin,the proliferation rates of PANC-1 cells were 0. 36% ± 0. 02%,0. 32% ± 0. 02% and 0. 26% ± 0. 01%,respectively. Significant difference was found between the control group 1 and the melatonin groups with concentrations of 1 000 and 1 000 000 nmol / L melatonin( all P < 0. 05). The proliferation rate of the control group 2 was 0. 43% ± 0. 02%,the proliferation rate in the DAPT group was 0. 30% ± 0. 02%( P < 0. 05). The expression of NICD1 in the control group 1 was 0. 47 ± 0. 05. After adding the concentrations of 1,1 000 and 1 000 000 nmol / L melatonin,the expression of NICD1 was 0. 32 ± 0. 04,0. 33 ± 0. 05 and 0. 21 ± 0. 03,respectively. Significant difference in the NICD1 expression was found between the control group 1 and the melatonin groups with concentrations of 1 000 and 1 000 000 nmol / L melatonin( all P < 0. 05). The expression of NICD1 in the control group 2 was 0. 63 ± 0. 05. The expression of NICD1 in the DAPT group was 0. 38 ± 0. 03( P <0. 05). Conclusion Melatonin can inhibit the proliferation of PANC-1 cells,whose mechanism may be involved in regulating the Notch signaling pathway.
