血管紧张素Ⅱ对小鼠胰岛β细胞miR-375表达的影响 被引量:2

摘要 目的观察血管紧张素Ⅱ(AngⅡ)对小鼠胰岛β细胞微小RNA-375(miR-375)表达的影响。方法将对数生长期的小鼠胰岛β细胞株MIN-6随机分为4组,A、B、C组分别加入终浓度为10-4、10-5、10-6mol/L的AngⅡ,D组加入等体积的PBS。收集各组培养24 h及B组培养24、48、72 h后的细胞,采用RT-PCR法检测miR-375。结果培养24 h后,A、B、C、D组miR-375相对表达量分别为3.75±0.01、3.13±0.23、1.12±0.17、1.00±0.00;A、B组与C、D组比较,P均<0.05;C组与D组比较,P>0.05。培养24、48、72 h后,B组miR-375相对表达量分别为3.13±0.23、2.84±0.02、2.80±0.01;B组各时点miR-375相对表达量两两比较,P均>0.05;而与D组比较,P均<0.05。结论 AngⅡ可上调小鼠胰岛β细胞中miR-375的表达。 Objective To observe the influence of AngiotensinⅡ( AngⅡ) on the microRNA( miR)-375 expression in the pancreatic β-cell of mice. Methods MIN-6 cells in logarithmic phase were divided into four groups: groups A,B,C and D. The cells in the groups A,B and C were treated with 10- 4mol / L AngⅡ,10- 5mol / L AngⅡ and 10- 6mol / L AngⅡ,respectively; and group D was treated with the same volume of PBS. Every group was treated for 24 hours to obtain the cells,and group B was treated for 24 hours,48 hours and 72 hours. The miR-375 expression was detected by RT-PCR.Results After being cultured for 24 h,the miR-375 expression in the groups A,B,C and D was 3. 75 ± 0. 01,3. 13 ±0. 23,1. 12 ± 0. 17 and 1. 00 ± 0. 00,respectively. When we compared groups A and B with groups C and D,statistical difference was found( all P < 0. 05). There was no statistical difference between group C and group D( P > 0. 05). After being cultured for 24 h,48 h and 72 h,the miR-375 expression in group B was 3. 13 ± 0. 23,2. 84 ± 0. 02 and 2. 80 ±0. 01. There was no statistical difference in the miR-375 expression at different time points of group B( all P > 0. 05).while,the miR-375 expression at different time points of group B was statistically different from that of group D( all P <0. 05). Conclusion AngⅡ may up-regulate the miR-375 expression in the pancreatic β-cell of mice.
