单核细胞趋化蛋白-1对人卵巢癌CaOV3细胞黏附、侵袭能力的影响及其机制 被引量:3

摘要 目的探讨单核细胞趋化蛋白-1(MCP-1)对人卵巢癌Ca OV3细胞黏附、侵袭能力的影响及其机制。方法将对数生长期的人卵巢癌Ca OV3细胞随机分为观察组和对照组,观察组经MCP-1(20μg/m L)预处理,对照组不予处理。两组行体外黏附试验,记录黏附Fibronectin、Collagen及Laminin蛋白的细胞数;行体内黏附试验,记录黏附的细胞球体数目、体积及部位;行细胞侵袭试验,记录穿膜细胞数。采用Western blotting法检测两组上皮-间质转化指标(上皮指标E-cadherin,间质指标Vimentin、N-cadherin)的表达。结果观察组黏附Fibronectin、Collagen及Laminin蛋白的细胞数及穿膜细胞数均高于对照组,P均<0.01。两组肿瘤细胞以细胞球体形式黏附在小鼠大网膜脂肪细胞部位,并且观察组黏附的细胞球体数目更多、球体更大。观察组E-cadherin低于对照组,Vimentin、Ncadherin均高于对照组,P均<0.01。结论 MCP-1可提高人卵巢癌Ca OV3细胞的黏附、侵袭能力,可能与其促进上皮细胞间质转化有关。
