摘要
目的探讨大蒜素对人宫颈癌He La细胞周期和凋亡的影响。方法将2.5、5、10、20、40、80、120μg/m L大蒜素分别作用于体外培养的He La细胞,24、48、72 h后,于倒置相差显微镜下观察He La细胞形态学变化;采用WST-8法测定He La细胞的增殖抑制率;流式细胞术测定He La细胞的凋亡率和细胞周期。结果倒置相差显微镜下观察到细胞凋亡的形态学改变。大蒜素干预后He La细胞增殖受抑,且呈浓度与时间依赖性(P均<0.05);随着大蒜素浓度的增加,细胞凋亡率增加,G1期细胞比例增加。结论大蒜素能阻滞He La细胞的周期进展,诱导细胞凋亡。
Objective To investigate the influence of allicin on the cell cycle progression and apoptosis of human cervical carcinoma He La cells. Methods The He La cells cultured in vitro were treated with different concentrations of allicin( 2. 5,5,10,20,40,80 and 120 μg/m L) for 24 h,48 h and 72 h. Then,the morphologic changes were observed by inverted phase contrast microscope. WST-8 was used to measure the proliferative inhibition rate of allicin on He La cells,and the cell cycle progression and apoptosis rate were detected by flow cytometry. Results The morphological changes of apoptosis was observed by inverted phase contrast microscope. The proliferation of He La cells was inhibited after the intervention of Allicin,which was in a dose- and time- dependent manner( all P < 0. 05). With the increasing concentration of allicin,the apoptosis rate was increased and the proportion of G0/ G1 cells was increased significantly. Conclusion Allicin can inhibit the He La cell proliferation,which may be related to the inducing cell apoptosis and blocking cell cycle progression.
