摘要
目的 :研究桂皮酸 (cinnamates ,CINN)诱导人急性早幼粒细胞白血病细胞系NB4 凋亡的能力 ,并探讨其机制。方法 :采用光镜形态学观察、凋亡细胞DNA片段原位末端标记法 (TUNEL)以及流式细胞术 (FCM)检测细胞凋亡 ;应用EnvisionTM免疫组化的方法检测凋亡抑制基因Bcl 2编码蛋白的表达。结果 :桂皮酸作用NB4 细胞后 ,出现典型的凋亡形态学改变 ;TUNEL标记也证实凋亡的发生 ;流式细胞仪检测有亚二倍体细胞峰的存在。在桂皮酸诱导NB4 细胞凋亡的过程中 ,Bcl 2基因蛋白表达水平逐渐下调。结论 :桂皮酸具有诱导NB4 细胞凋亡的作用 ;Bcl 2基因表达水平的下调 ,可能是NB4 细胞走向凋亡的重要条件。
Objective: To study the apoptosis of NB 4 promyelocytic leukemia cells induced by CINN and to illustrate the possible mechanism. Methods: Apoptosis was detected by morphological observation (light microscopy),TUNEL labeling method and flow cytometry. Bcl 2 expression was analysed by immunocytochemistry. Results: NB 4 cells treated with CINN exhibited a morphological characteristic of apoptosis. TUNEL labeling confirmed the present of apoptosis cells. FCM analysis showed that there was Sub G1 cells. Bcl 2 was downregulated during the apoptosis process. Conclusion: CINN can induce apoptosis of NB 4 cells. Downregulation of Bcl 2 expression appears to play an important role in the apoptosis of leukemic cells.
