应用Sj23基因重组抗原诊断牛、羊血吸虫病研究 被引量:6

摘要 以血吸虫基因重组抗原LHD Sj23/pGEX(血吸虫23kD抗原大亲水区多肽/pGEX载体表达蛋白)作为抗原,应用ELISA法检测黄、水牛和绵羊血吸虫病。结果对黄、水牛的阳性符合率为94 0%(79/84)和85 5%(53/62),阴性符合率为82 7%(81/98)和95 1%(77/81),对绵羊的阳性符合率为86 04%(111/129),阴性符合率为100%(91/91)。以血吸虫成虫抗原(SWAP)及虫卵抗原(SEA)作为诊断抗原获得的结果差异不明显。对锥虫感染绵羊血清不出现交叉反应。由于基因重组抗原的制备比虫体抗原制备简便、价廉,同时由于基因重组抗原的应用有利于诊断技术的标准化,以基因重组抗原作为血吸虫病诊断抗原具有良好的研究应用前景。 The fusion protein LHDSj23/pGEX was employed as diagnosing antigen for detecting bovine and sheep schistosomiasis by ELISA.The result showed that 94.0%(79/84),85.5%(53/62)and 86.04%(111/129)positive coincidence rates,82.7%(81/98),95.1%(77/81)and 100%(91/91)negative coincidence rates were obtained from yellow cattle,water buffaloes and sheep respectively.The similar results were obtained by using SWAP,SEA as diagnosing antigen comparing with that of recombinant LHDSj23/pGEX.No cross reaction was found in 24 sheep Trypanosomosis. The study showing that using Sj23 recombinant antigen as the diagnosis reagent for schistosomiasis will have good potential for applying in the field.
