基于两种一测多评法对尾叶香茶菜中四种二萜类成分的质量控制研究 被引量:1

摘要 目的:建立同时测定尾叶香茶菜中Kamebakaurin、excisanin A、rabdokunmin C、Kamebanin四种二萜类成分的HPLC方法,在此基础上,建立一测多评法方法学考察模式,以验证一测多评法在尾叶香茶菜药材质量控制中应用的可行性和技术适用性。方法:以尾叶香茶菜中的4种活性成分为指标成分,采用两种校正方法分别建立各成分与Kamebakaurin的相对校正因子(fk/s),计算各成分含量,实现一测多评;同时采用外标法测定尾叶香茶菜中这4种成分的含量,并比较两种一测多评法所得计算值与实测值的差异,以验证一测多评法的准确性和可行性。结果:建立了尾叶香茶菜中4种二萜类成分一测多评法方法学考察模式;不同仪器和色谱柱多点校正法的计算值与外标法实测值之间比较差异无统计学意义,斜率校正法的计算值与外标法实测值之间比较差异有统计学意义。结论:同时测定尾叶香茶菜中Kamebakaurin、excisanin A、rabdokunmin C、Kamebanin 4种二萜类成分的多点校正法准确可靠,可用于尾叶香茶菜药材的质量控制。 Objective:To establish a HPLC method to determine four active components(Kamebakaurin,excisanin A,rabdokunmin C and Kamebanin)in Rabdosia excisa(Maxim.). The methodological investigation mode of QAMS was established to verify the feasibility and technical adaptability of this approach in quality control of Rabdosia excisa. Methods:Four components were used as index components,two correction methods were used to establish the relative correction factor(fk/s)between each component and Kamebakaurin. Then fk/s was used to calculate the content of each component in Rabdosia excisa. Meanwhile external standard method was used to determine the content of the four compounds and the difference between calculated value and measured value was compared to verify the accuracy and feasibility of QAMS.Results:The methodological investigation mode of QAMS which was used to evaluate the four kinds of diterpenoids of Rabdosia excisa was established. There was not significant difference between the value calculated by multipoint correction with different instruments and columns and the values measured by external standard method. But there was significant difference between the value calculated by slope correction and the values measured by external standard method. Conclusion:Multipoint correction for calculating the content of Kamebakaurin,excisanin A,rabdokunmin C and Kamebanin in Rabdosia excisa is accurate and reliable,it could be used to control the quality of Rabdosia excise.
