摘要
为检测皮肌炎(dermatomyositis,DM)血清微小RNA(microRNA,miR)-150-5p的表达水平,探讨其与肌炎特异性自身抗体(myositis specific autoantibody, MSA)和不同临床表型的相关性,收集2011年3月至2016年2月在中日友好医院风湿免疫科住院治疗的DM患者血清49例,健康对照者血清30例,提取血清总RNA,采用实时荧光定量PCR检测血清miR-150-5p的相对表达水平,分析其与MSA和不同临床表型的相关性。结果显示,DM组患者血清miR-150-5p水平[4.37(2.49, 7.11)]低于健康对照组[10.67(1.59, 34.66),P=0.002],抗MDA5抗体阳性患者血清miR-150-5p水平(3.58±2.02)低于抗MDA5抗体阴性患者(6.19±5.27,P=0.017);抗核基质蛋白2(nuclear matrix protein 2, NXP2)抗体阳性患者血清miR-150-5p水平(2.89±2.00)低于抗NXP2抗体阴性患者(5.99±5.02,P=0.047)。不同间质性肺疾病(interstitial lung disease,ILD)组织学分型患者间血清miR-150-5p水平差异具有统计学意义(P<0.05)。提示miR-150-5p可能参与了DM自身抗体的致病过程。
This study was designed to detect the expression of serum microRNA(miR)-150-5 p in dermatomyositis(DM)and to explore its association with myositis specific autoantibodies(MSA)and different clinical phenotypes.Forty-nine samples of serum from hospitalized patients with DM were collected.Thirty healthy serum samples served as controls.Total RNA was extracted from serum.The expression level of miR-150-5 p was detected by real-time fluorescent quantitative PCR and analyzed for its correlation with MSA and different clinical phenotypes.The results showed that the relative expression of serum miR-150-5 p in DM patients[4.37(2.49,7.11)]was lower than that in healthy controls(10.67(1.59,34.66),P=0.002)Serum miR-150-5 p level in anti-MDA5 antibody-positive patients(3.58±2.02)was lower than that in anti-MDA5 antibody-negative patients(6.19±5.27,P=0.017).Moreover,anti-nuclear matrix protein 2(NXP2)antibody-positive patients(2.89±2.00)had lower serum miR-150-5 p levels than anti-NXP2 antibody-negative patients(5.99±5.02,P=0.047).Finally,serum miR-150-5 p showed different expression levels among different histological types of interstitial lung disease(ILD,P<0.05).Altogether,these results suggest that miR-150-5 p may be involved in the production of DM autoantibodies.
