北五味子不同活性部位对大鼠酒精性肝损害的影响及对品质鉴别的意义

Effect of active ingredients of fructus schisandrae chinensis on rats with Alcoholic liver damage

目的探索北五味子不同活性部位对酒精引起的大鼠肝损伤的影响。方法取Wistar大鼠125只,随机分为对照组、损伤组、五味子木质素组、多糖组、挥发油组,用酒精复制急性酒精中毒肝损伤模型。提取北五味子中的主要成分,之后用北五味子活性成分木质素、多糖、挥发油给各组大鼠灌胃。检测各组血清中ALT、AST活性,肝匀浆MDA、SOD、ATPase、γ-谷氨酰转移酶(γ-GT)活性及含量,光镜下观察肝脏结构变化。最后检测各提取物对肿瘤坏死因子-α(TNF-α)的影响。结果木质素组、多糖组大鼠血清中ALT、AST活性及肝匀浆中MDA较模型组显著降低(P<0.05),挥发油组无明显变化。木质素组、多糖组肝脏匀浆中SOD、ATPase、γ-GT酶活性较模型组显著增强(P<0.05)。挥发油可增强SOD、ATPase活性但不能增强γ-GT酶活性。光镜下观察木质素组、多糖组大鼠组织脂肪变、线粒体肿胀的有所缓解。木质素、多糖可抑制TNF-α的释放。结论北五味子活性成分木质素、多糖对酒精引起的肝脏损伤有保护作用,可作为相应的评价指标。

Objective To investigate the effect of active ingredients of fructus schisandrae chinensis on rats with alcoholic liver damage. Methods125 Wistar rats were randomly divided into control group,injury group,lignans group,polysaccharide group and volatile oil group,and acute alcoholic liver injury models were constructed. The main ingredient of fructus schisandrae chinensis were extracted and lavage the rats with lignin,polysaccharides,volatile oil of fructus schisandrae chinensis,the activity of serum ALT and AST,liver homogenate MDA,SOD and ATPase,gamma glutamine transferase( gamma GT) activity and its content in each groups were detected,the change of liver structure were observed and various extracts on the tumor necrosis factor alpha( TNF alpha) were detected. Results Rats serum ALT,AST activitives and MDA contents in lignin group and polysaccharide group were significantly lower than model group( P < 0. 05). There was no significant alteration of the volatile oil. The enzyme activity of SOD,ATPase,γ-GT in liver homogenate of lignin group and polysaccharide group were significantly enhanced( P < 0. 05). Volatile oil increased SOD and ATPase activity but not enhanced γ-GT enzyme activity. Then observe the lignin fatty change caused by alcohol,polysaccharide group organization,mitochondrial swelling eased. Lignin,polysaccharide can inhibit the release of TNF-α. Conclusion Lignin,polysaccharide,the active ingredients of fructus schisandrae chinensis,protects liver damage caused by alcohol.
