蚤休提取物对实验性脑出血大鼠脑损伤的保护作用研究 被引量:1

摘要 目的探讨蚤休提取物对实验性脑出血大鼠脑损伤的保护作用及机制。方法 100只健康SD大鼠依据随机数字表法随机分为5组,假手术组、模型组和蚤休提取物低、中、高剂量组(浓度分别为25、50、100 mg/kg),每组各20只。注射Ⅶ型胶原酶建立大鼠脑出血模型,蚤休提取物组给于相应浓度灌胃,模型组和假手术组给于生理盐水。分别于灌胃后第3、5、7天对各组大鼠进行神经功能缺损评分,并每组取6只大鼠进行脑组织含水量、脑组织中超氧化物歧化酶(SOD)含量和血浆中TNF-α含量检测。结果与假手术组相比,模型组大鼠神经功能缺损评分显著升高(P<0.01);与模型组相比,蚤休提取物低、中、高剂量组均能显著降低大鼠神经功能缺损评分(P<0.05,P<0.01)。与假手术组相比,模型组大鼠脑组织SOD活性显著降低(P<0.01);与模型组相比,蚤休提取物中、高剂量组均能显著降低大鼠神经功能缺损评分(P<0.05)。与假手术组相比,模型组大鼠各时间点血浆中TNF-α含量显著升高(P<0.01);与模型组相比,蚤休提取物中、高剂量组均能显著降低大鼠血浆中TNF-α含量(P<0.05,P<0.01)。结论蚤休提取物能够降低Ⅶ型胶原酶诱导的脑出血大鼠的神经功能缺损评分,改善脑组织水肿症状,通过提高脑组织血浆SOD水平,抑制脂质体氧化,减少TNF-α的产生,对脑出血大鼠受损的脑组织具有保护作用。 Objective To explore the protective effect and mechanism of the extract of Paris polyphylla on hemorrhagic brain injury in rats.Methods 100 healthy SD rats were randomly divided into sham operative group( n = 20),cerebral hemorrhage model group( n = 20) and the extract of Paris polyp- hylla treatment group( three dirrerent doses at 25,50,100 mg / kg with 20 rats in each group) according to random number table. Cerebral hemorrhage rats model induced by Ⅶ collagenase was established. ICH model group and the sham group was given saline,and the treatment group was given different doses of Paris polyphylla extract. The neurological deficit scores of each group were evaluated after treatment at the 3rd,5th,7thday. Then six rats were selected and killed in each group at different time point. The level of brain water and SOD in hemorrhage brain tissue were detected,so were the content of TNF-α in plasma. Results The neurological deficit scores in model group significantly increased compared with sham group with statistically significant difference( P < 0. 01); compared with model group,groups of low dose,middle dose and high dose significantly decreased neurological deficit scores( P < 0. 05,P < 0. 01). SOD activity levels in brain tissue of model group significantly decreased compared with sham group( P < 0. 05); the neurological deficit scores in groups of middle dose and high dose significantly decreased compared with model group with statistically significant difference( P < 0. 05). The level of TNF-α in plasma of the model group significantly increased compared with sham group at each time point with statistically significant difference( P < 0. 01). The level of TNF-α in plasma of groups of middle dose and high dose significantly decreased compared with model group( P < 0. 05,P < 0. 01). Conclusion The extract could reduce the scores of neurological deficit scores of collagenase-induced intracerebral hemorrhage in rats,improve the symptoms of cerebral edema,increase the SOD levels in brain tissue,inhibit the oxidation of liposomes,reduce inflammation cytokine of TNF-α,and protect the damaged brain tissue in cerebral hemorrhage rats.
