益气升阳方调节apoE(-/-)动脉粥样硬化小鼠脂质代谢及其抗氧化和抗炎作用研究 被引量:2

Antioxidation and Anti-inflammatory Effects of“Yiqi Shengyang Decoction”and its Regulatory Effect on Lipid Metabolism in apo E(-/-) Atherosclerosis Mice

摘要 目的:探讨益气升阳方调节apo E(-/-)动脉粥样硬化(AS)模型小鼠脂质代谢及其抗氧化、抗炎作用。方法:将32只6~8周apo E(-/-)小鼠高脂喂养12周后制成AS动物模型,随机分为益气升阳方药组(复方组)、黄芪组、西药(洛伐他汀)组、模型组,每组8只;同时,高脂饲养8只同一品系C57小鼠设为正常组。干预组分别给予益气升阳方药、黄芪和洛伐他汀,均采用灌胃给药,同时继续高脂饮食;正常组和模型组小鼠持续高脂饮食饲养。干预12周。采用酶偶联比色法检测各组小鼠血清TG、TC含量;TBA法及羟胺法分别检测MDA、SOD含量;选择测定法检测血清LDL-C、HDL-C含量;ELISA法检测MCP-1、TNF-α表达。结果:模型组小鼠血清TC、LDL-C、MDA水平及MCP-1、TNF-α表达较正常组均有显著升高(P<0.05),血清HDL-C水平、SOD活性较正常组明显下降(P<0.05);通过益气升阳方药干预后小鼠血清TC、LDL-C、MDA、SOD、TNF-α变化较模型组均有明显改善(P<0.05),但与模型组相比,该复方不能明显改变HDL-C、MCP-1含量。结论:益气升阳方药和黄芪均可改善动脉粥样硬化动物的脂质代谢,并具有抗氧化及抗炎作用。 Objective: To explore the antioxidation and anti-inflammatory effects of'Yiqi Shengyang Decoction'and its regulatory effect on lipid metabolism in mice. Methods: Thirty-two apo E(-/-) mice aged 6-8 weeks were fed with high fat diet for 12 weeks and randomly divided into the'Yiqi Shengyang Decoction'group,Radix Astragali seu Hedysari group,western medicine group( lovastatin) and model control group,with 8 mice in each group; the same strain C57 mice with high-fat feeding were set as the normal group. Intervention groups were treated by 'Yiqi Shengyang Decoction',Radix Astragali seu Hedysari and lovastatin by lavage administration and feed on high fat diet at the same time. The mice of the normal group and the model control group were fed on high fat diet for 12 weeks. The contents of TG and TC in the serum were detected by colorimetric method; the level of MDA and SOD were detected by TBA and hydroxylamine method respectively; the level of LDL-C and HDL-C in serum were detected by selective measuring method; expression of MCP-1 and TNF-α were detected by ELISA method. Results: Compared with the normal group,the levels of TC,LDL-C and MDA and the expression of MCP-1and TNF-α in the model group were increased markedly( P < 0. 05),while the HDL-C level and SOD activity were reduced( P < 0. 05); compared with the model control group,the levels of TC,LDL-C,MDA,SOD and TNF-α in serum were improved after intervened by'Yiqi Shengyang Decoction'( P < 0. 05),but compared with the model group,no significantly change was found in the levels of HDL-C and MCP-1. Conclusion: 'Yiqi Shengyang Decoction 'and Radix Astragali seu Hedysari can improve the metabolism in atherosclerotic animals,with its antioxidant and anti-inflammatory effects.
