摘要
目的:探讨骨髓间充质干细胞(BMSCs)联合柴胡疏肝散对心肌梗死大鼠心肌组织的保护作用。方法:采用全骨髓贴壁法培养BMSCs。经结扎冠状动脉制备大鼠心肌梗死(AMI)模型。SD大鼠随机分为假手术组、AMI组(模型组)、BMSCs组、CHSG-L组(柴胡疏肝散小剂量+BMSCs移植)、CHSG-M组(柴胡疏肝散中剂量+BMSCs移植)、CHSG-H组(柴胡疏肝散大剂量+BMSCs移植)。BMSCs细胞悬液直接注入梗死区边缘心肌组织;中药或生理盐水经灌胃给予。各组干预7 d后,生化法测定大鼠血清超氧化物歧化酶(SOD)与丙二醛(MDA)含量;ELISA法检测大鼠血清低氧诱导因子(HIF-1α)表达;免疫组化法观察心肌血管内皮生长因子(VEGF)蛋白表达。结果:与模型组和BMSCs组比较,各联合治疗组SOD升高、MDA降低,血清HIF-1α水平和心肌细胞VEGF蛋白表达升高;且柴胡疏肝散剂量越大,作用越显著。结论:BMSCs移植联合柴胡疏肝散可一定程度上保护心肌梗死大鼠心肌组织。
Objective: To discuss'Chaihu Shugan Powder'combined with BMSCs transplantation's protective effect on the myocardium of the AMI rats. Methods: BMSCs were cultivated by bone marrow adherent method. Rats were modeled as AMI by coronary artery ligation. SD rats were randomly divided into 6 groups: sham group,AMI group,BMSCs group,CHSGL group( low concentration decoction of'Chaihu Shugan Powder'by intragastric administration and BMSCs transplantation),CHSG-M group( middle concentration decoction of 'Chaihu Shugan Powder ' by intragastric administration and BMSCs transplantation),CHSG-H group( high concentration decoction of'Chaihu Shugan Powder'by intragastric administration and BMSCs transplantation). BMSCs cell suspension were injected into myocardium tissue in AMI area; Chinese medicine or normal saline were administered by gavage. 7 days later,the rats' serum was used to detect SOD and MDA by the biochemical method,while detect HIF-1α by ELISA method. The myocardial tissue at the edge of the myocardial infarction area was used to detect VEGF by immunohistochemical method. Results: Compared with the model group and BMSCs group,all treatment groups could increase the content of SOD while decrease the content of MDA,and increase the content of HIF-1α in rats' serum,and increase the protein expression of VEGF in myocardial cells. The effect was enhanced with the increase of concentration of'Chaihu Shugan Powder'. Conclusion: 'Chaihu Shugan Powder'combined with BMSCs transplantation could protect the myocardium of the AMI rats.
