冠心康含药血清对RAW264.7巨噬细胞MerTK表达的影响 被引量:2

摘要 目的:通过观察冠心康含药血清对RAW264.7巨噬细胞Mer受体酪氨酸激酶(Mer tyrosine kinase,Mer TK)表达的影响,探讨冠心康抗动脉粥样硬化的可能作用机制。方法:SD大鼠给予冠心康灌胃,2次/d,连续给药5 d后制备冠心康含药血清。体外常规培养RAW264.7巨噬细胞,实验分为空白对照组(加10%浓度的正常血清)、模型组[加10%浓度的正常血清+20 mg/L氧化型低密度脂蛋白(ox-LDL)]、冠心康组(加10%浓度的冠心康含药血清+20 mg/L ox-LDL)。干预12 h后采用RT-q PCR检测各组巨噬细胞的Mer TK mRNA表达;干预24 h后采用Western blot法检测各组巨噬细胞的Mer TK蛋白表达。结果:与空白对照组比较,模型组Mer TK mRNA和蛋白的表达水平明显降低(P<0.01);与模型组比较,冠心康组Mer TK mRNA和蛋白的表达水平均明显升高(P<0.01)。结论:冠心康抗动脉粥样硬化的作用可能与上调巨噬细胞Mer TK的mRNA和蛋白表达有关。 Objective: To observe the effects of Guanxinkang medicated serum on Mer tyrosine kinase( Mer TK) expression in RAW264.7 macrophages,and discuss the possible mechanism of Guanxinkang on atherosclerosis. Methods: SD rats were treated with Guanxinkang decoction by intragastric administration twice a day for 5 days to prepare the medicated serum. The RAW264.7 macrophages were cultured in vitro. The cells were divided into the blank control group( treated with 10% normal serum),model group( treated with10% normal serum and 20 mg/L ox-LDL) and Guanxinkang group( treated with 10% medicated serum and 20 mg/L ox-LDL). The mRNA expression of Mer TK in macrophages was detected by RT-q PCR after intervention for 12 hours,and the protein expression of Mer TK in macrophages was detected by Western blot after intervention for 24 hours. Results: Compared with the control group,the mRNA and protein expression levels of Mer TK were obviously decreased in the model group( P<0.01). Compared with the model group,the mRNA and protein expression levels of Mer TK were obviously increased in the Guanxinkang group( P<0.01). Conclusion: The mechanism of Guanxinkang against atherosclerosis may be associated with the up-regulation of Mer TK mRNA and protein expressions in macrophages.
