滋阴固本颗粒对实验性自身免疫性脑脊髓炎大鼠视神经MBP表达的影响 被引量:1

Effects of Ziyin Guben Granule on expression of myelin basic protein in optic nerve of rats with experimental autoimmune encephalomyelitis

摘要 目的:探讨滋阴固本颗粒对实验性自身免疫性脑脊髓炎(EAE)大鼠视神经碱性髓鞘蛋白(MBP)表达的影响。方法:雌性Wistar大鼠随机分成正常组、模型组、激素组及滋阴固本颗粒(中药)低、中、高剂量组。除正常组外,均用豚鼠脊髓匀浆(GPSCH)+完全福氏佐剂(CFA)+百日咳疫苗(PV)致敏雌性Wistar大鼠,建立EAE模型。造模后次日即给予相应干预,激素组给予醋酸地塞米松6.25 mg·kg^(-1)·d^(-1),中药低、中、高剂量组分别给予中药液13.33 g·kg^(-1)·d^(-1)、26.67 g·kg^(-1)·d^(-1)、53.33 g·kg^(-1)·d^(-1),正常组和模型组给予等容量0.9%Na Cl溶液,1次/d,连续灌胃14 d。干预结束后对大鼠进行神经功能评分,免疫组化法检测视神经组织内MBP表达情况。结果:与正常组比较,模型组神经功能评分明显升高(P<0.01);与模型组比较,激素组与中药中、高剂量组大鼠的神经功能评分均明显降低(P<0.01)。与正常组比较,模型组视神经MBP表达明显降低(P<0.01),激素组和中药低、中、高剂量组MBP表达水平均明显高于模型组(P<0.01);激素组与中药高剂量组MBP表达差异无统计学意义(P>0.05),中药中、高剂量组MBP表达高于中药低剂量组(P<0.01)。结论:滋阴固本颗粒对EAE大鼠具有防治作用,其作用可能与其增强视神经组织内MBP的表达有关。 Objective: To discuss the effects of Ziyin Guben Granule on expression of myelin basic protein( MBP) in optic nerve of rats with experimental autoimmune encephalomyelitis( EAE). Methods: The female Wistar rats were sensitized by guinea pig spinal cord homogenate and complete Freund's adjuvant and pertussis vaccine to establish the EAE model. The female Wistar rats were randomly divided into the normal group,model group,hormone group and Ziyin Guben Granule groups with low-,medium-and high-dose. Except the normal group,the model was established in the other groups. After modeling,each group was treated with the corresponding drugs. The hormone group was treated with dexamethasone acetate at daily dose of 6.25 mg/kg,the Ziyin Guben Granule groups with low-,mediumand high-dose were treated with Ziyin Guben Granule decoction at daily dose of 13. 33,26. 67 and 53. 33 g/kg respectively,the normal group and model group were treated with 0. 9% Na Cl solution at equivalent volume. The administration was performed once a day for continuous 14 days. After intervention,the neurological function score was assessed in rats. The expression of MBP in the optic nerve tissue was detected by immunohistochemistry. Results: Compared with the normal group,the neurological function score was significantly increased in the model group( P<0.01). Compared with the model group,the neurological function score was obviously decreased in the hormone group and Ziyin Guben Granule groups with medium-and high-dose( P < 0. 01). Compared with the normal group,the MBP expression was obviously decreased in the model group( P < 0.01). The level of MBP expression in the hormone group and Ziyin Guben Granule groups with low-,medium-and high-dose was obviously higher than that in the model group( P<0.01). There was no statistically significant difference on the MBP expression between the hormone group and Ziyin Guben Granule group with high-dose( P>0.05). The MBP expression in the Ziyin Guben Granule groups with medium-and high-dose was higher than that in the low-dose group( P< 0.01).Conclusion: Ziyin Guben Granule shows the preventive and therapeutic effects on EAE rats. Its mechanism may be associated with the upregulation of MBP expression in the optic nerve tissue.
