摘要
目的探讨三七总皂苷(PNS)对乙酰胆碱(ACh)-氯化钙(Ca Cl_2)诱导的大鼠心房颤动(简称房颤)及期前收缩的干预作用。方法ACh-Ca Cl_2混合液为造模药物,以尾静脉注射的方式建立房颤动物模型。经过监测24 h动态心电图,筛选出具有典型房颤心电图表现的大鼠,随机分为正常组、模型组、PNS组。各组给予相应的干预措施。各组均采用24 h动态心电图监测,直到实验结束。采用苏木素-伊红(HE)染色、三色(Masson’s thrichrome)染色,光学显微镜观察心脏形态学改变;采用免疫酶联吸附法(ELISA)检测大鼠血清IL-17A水平。结果(1)与正常组比较,模型组大鼠房颤持续时间显著增加(P<0.05);与模型组比较,PNS组大鼠房颤持续时间显著减少(P<0.05)。(2)与正常组比较,模型组大鼠期前收缩发生率显著升高(P<0.05);与模型组比较,PNS组大鼠期前收缩率显著降低(P<0.05)。(3)与正常组比较,模型组心肌有大量炎性细胞浸润,心肌局部呈灶性坏死,有大量胶原纤维生成;与模型组比较,PNS组心肌细胞排列相对整齐,仅在血管周围见少量炎性细胞浸润和胶原沉积(P<0.05)。(4)与正常组比较,模型组血清IL-17A水平显著上升(P<0.05);与模型组比较,PNS组血清IL-17A水平显著下降(P<0.05)。结论PNS能够显著抑制由ACh-Ca Cl_2混合液诱发的大鼠房颤以及期前收缩等心律失常,并能显著减轻房颤诱发的心肌纤维化,其可通过抑制炎症反应进而抑制房颤。
Objective To investigate the intervention effects of Panax Notoginseng Saponins(PNS)on ACh-Ca Cl_2 induced atrial fibrillation and proiosystole in rats.Methods Animal model of atrial fibrillation was established by acetylcholine(ACh)-calcium chloride(Ca Cl_2)mixture for tail intravenous injection.Afte monitoring dynamic electrocardiogram for 24 hours,the rats with typical electrocardiogram of atrial fibrillation were screened out,and then randomly divided int the normal group,model group and PNS group.Each group was received the corresponding intervention.The dynamic electrocardiogram monitoring for 24 hour was applied in each group until the end of the experiment.The morphological changes of heart were observed by light microscope after hematoxylin and eosin(HE)and Masson’s thrichrome staining.The serum level of IL-17A was detected by enzyme linked immunosorbent assay(ELISA).Results(1)Compared with th normal group,the duration of atrial fibrillation in the model group was significantly increased(P<0.05).Compared with the model group,the duration of atria fibrillation in the PNS group was significantly decreased(P<0.05).(2)Compared with the normal group,the proiosystole incidence in the model group wa significantly increased(P<0.05).Compared with the model group,the proiosystole incidence in the PNS group was significantly decreased(P<0.05)(3)Compared with the normal group,a large number of inflammatory cells infiltration and local necrosis in the myocardium and many collagen fibers were observed in the model group.Compared with the model group,the myocardial cells in the PNS group arranged in order,only a small amount of inflammatory cell infiltration and collagen deposition were found around the vessels(P<0.05).(4)Compared with the normal group,the serum level of IL-17A in the model group wa significantly increased(P<0.05).Compared with the model group,the serum level of IL-17A in the PNS group was significantly decreased(P<0.05)Conclusion PNS can significantly inhibit the atrial fibrillation and proiosystole induced by ACh-Ca Cl_2 in rats,and relieve the myocardial fibrosis induced by atria fibrillation.Its mechanism may relate to the inhibition of inflammatory reaction.
