鹿角脱盘5种提取部位对H_(22)荷瘤小鼠免疫功能的影响 被引量:2

摘要 目的评价鹿角脱盘5种提取部位对肝癌H_(22)荷瘤小鼠免疫功能的影响。方法将90只SPF级昆明种小鼠随机分成正常阴性对照组、模型组、阳性对照组、鹿角脱盘酸(盐酸)、碱(氢氧化钠)、盐(氯化钠)、醇(乙醇)和水5种提取部位组和鹿角脱盘原粉组共9组,每组10只。分别以脾脏组织病理变化、外周血和脾脏CD4^+和CD8^+T淋巴细胞百分比、血清中细胞因子IL-6、IL-12和抗体IgE、IgG、IgM水平为指标,评价鹿角脱盘的盐酸、氢氧化钠、氯化钠、乙醇和水5种提取部位提高荷瘤小鼠免疫功能的作用。结果与模型组比较,鹿角脱盘5种提取部位能够保护荷瘤小鼠脾脏结构;提高脾脏和外周血CD4^+/CD8^+比值(P<0.05);提高荷瘤小鼠血清中细胞因子IL-6、IL-12和抗体IgE、IgG、IgM的水平(P<0.05)。结论鹿角脱盘碱溶液提取部位效果最好;鹿角脱盘可能通过提高荷瘤小鼠体液免疫和细胞免疫的功能,逆转肿瘤发生发展对机体免疫功能的抑制作用,发挥抗肿瘤作用。 Objective To evaluate the effects of five kinds extracts of deer antler base on the immunological function in H_(22) tumor bearing mice. Methods A total of 90 SPF Kunming mice were randomly divided into the normal control group,model group,positive control group,group of original powder of deer antler base and groups of 5 kinds extracts of deer antler base that treated with acid,base,salt,alcohol and water respectively,10 mice in each group. By the pathological changes of spleen,the percentage of T-lymphocyte subsets CD4^+and CD8^+in peripheral blood and spleen and the serum levels of cytokines( IL-6,IL-12) and antibody( IgE,IgG,IgM),the effects of five kinds extracts of deer antler base which extracted by hydrochloric acid,sodium hydroxide,sodium chloride,ethanol and water on the immunological function in tumor bearing mice were evaluated. Results Compared with the model group,the 5 kinds extracts of deer antler base could protect the spleen structure of tumor-bearing mice,improve the ratio of CD4^+/CD8^+in spleen and peripheral blood( P < 0. 05),and increase the serum levels of IL-6,IL-12 and IgE,IgG,IgM( P < 0. 05). Conclusion The alkaline solution extract of deer antler base shows the best effects. Deer antler base may play the antitumor role by improving the humoral and cellular immunity of tumor bearing mice and reversing the inhibition on immunological function caused by tumor.
