摘要
目的通过观察鹿红方干预对充血性心力衰竭(CHF)大鼠心肌高能磷酸盐、心功能和心肌细胞病理重构的影响,筛选鹿红方有效成分。方法分离鹿红方全方水煎液,分别用蒸馏水、30%乙醇、60%乙醇和95%乙醇洗脱,得到A、B、C、D 4种提取液,根据正交实验分为A组、B组、AB组、C组、AC组、BC组、ABC组、D组、AD组、BD组、ABD组、CD组、ACD组、BCD组,第15组为曲美他嗪组,第16组为假手术组,每组6只。通过冠脉结扎法制备CHF大鼠模型,干预8周之后记录心功能变化,包括左室收缩压(LVSP)、左室舒张末压(LVEDP)、左室内压最大上升和下降速率;检测心肌组织ATP和ADP含量;心肌组织进行HE、VG染色,检测心肌胶原容积分数(ICVF)和心肌细胞横径(TMD)比值,观察心肌结构重建。结果与假手术组比较,鹿红方第7、8、12、13组及第15组可有效改善心力衰竭大鼠心功能;鹿红方第7组及第15组改善心肌磷酸腺苷指标;鹿红方第7、12组及第15组ICVF/TMD比值与假手术组相同(P>0.05),证明鹿红方第7、12组及第15组可以改善心力衰竭大鼠心肌结构重建。在改善心力衰竭心功能、心肌代谢紊乱及心肌结构重建三方面,第7组是鹿红方最佳有效成分。结论鹿红方能改善CHF大鼠能量代谢、心肌结构重建,从而改善心功能;鹿红方第7组(蒸洗液、30%液、60%液混合提取组)是最佳有效成分。
Objective To screen the effective components of Luhong Recipe by observing the effects of Luhong Recipe on myocardial high-energy phosphate,cardiac function and pathological remodeling of cardiomyocytes in rats with congestive heart failure( CHF). Methods The decoction of Luhong Recipe was separated by elution of distilled water,30% ethanol,60% ethanol and 95% ethanol respectively,and 4 kinds of extract were obtained and labeled as A,B,C and D solution. According to the orthogonal experiment,the objects were divided into group A,group B,group AB,group C,group AC,group BC,group ABC,group D,group AD,group BD,group ABD,group CD,group ACD,group BCD,trimetazidine group and sham operation group,which labeled from No. 1 to No. 16 group in turn,6 rats in each group. CHF rat model was established by coronary artery ligation,and the heart functions were observed after 8-week treatment,including the left ventricular systolic pressure( LVSP),left ventricular end-diastolic pressure( LVEDP) and maximum ascending and descending rate of left ventricle pressure( ±dp/dtmax). The contents of ATP and ADP in myocardial tissues were detected,and the ratio of myocardial interstitial collagen volume fraction( ICVF) and transverse myocardium diameter( TMD) was measured to observe the myocardial remodeling after haematoxylin-eosin( HE) staining and van-gieson( VG) staining.Results Compared with the sham operation group,the cardiac function of heart failure rats were significantly improved in No.7 group( group ABC),No.8 group( group D),No.12 group( group CD),No.13 group( group ACD) of Luhong Recipe and No.15 group( trimetazidine group). The levels of adenosine triphosphate were ameliorated in No.7 group( group ABC) and No.15 group( trimetazidine group). The ratio of ICVF/TMD in No.7 group( group ABC),No.12 group( group CD) and No.15 group( trimetazidine group) was close to that in the sham operation group( P > 0.05),which suggested that No.7 group,No. 12 group and No. 15 group of Luhong Recipe alleviated the myocardial remodeling of rats with heart failure. For the effects on improving cardiac function in heart failure,myocardial metabolic disorders and myocardial remodeling,No.7 group( group ABC) was the best effective component of Luhong Recipe. Conclusion Luhong Recipe can promote cardiac function by improving energy metabolism and myocardial remodeling,and No.7 group( distilled water,30% ethanol and 60% ethanol mixed extract group) is the best effective component.
