摘要
目的探讨电针对胰岛素抵抗型多囊卵巢综合征的影响。方法以来曲唑联合高脂饲料诱导胰岛素抵抗型多囊卵巢综合征大鼠模型。造模成功的大鼠随机分为模型组和电针组,并设同期普通饲料喂养的大鼠为对照组。电针组大鼠每日给予电针治疗(取"关元""中极"和双侧"三阴交"穴),连续4周。分别于治疗前后记录大鼠体质量,观察大鼠卵巢组织的病理学变化,检测大鼠血清睾酮(T)、空腹胰岛素(FINS),计算胰岛素敏感指数(ISI)。结果 HE染色后观察对照组大鼠可见多个不同发育阶段的卵泡,颗粒细胞呈多层;模型组大鼠卵巢组织多个卵泡囊性扩张,颗粒细胞层少,白膜增厚;电针组大鼠卵巢组织中囊性卵泡减少,颗粒细胞层增多。与对照组相比,模型组体质量、血清T和FINS水平显著升高,ISI显著下降(P<0.05,P<0.01);与模型组相比,电针组体质量、血清T水平显著降低(P<0.05),FINS水平降低,ISI升高,但无显著性差异。结论电针可降低胰岛素抵抗型多囊卵巢综合征大鼠的体质量和血清睾酮水平,在一定程度上改善胰岛素抵抗。
Objective To discuss the effects of electroacupuncture on rats of polycystic ovary syndrome with insulin resistance( PCOS-IR). Methods The rat model of PCOS-IR was established by letrozole combined with high fat diet. The successful modeling rats were randomly divided into the model group and electroacupuncture group,the rats fed with the normal diet in the same period were taken as the control group. The electroacupuncture group was treated with electroacupuncture at acupoints of Guanyuan( CV4),Zhongji( CV3) and bilateral Sanyinjiao( SP6) everyday,with a course of 4 weeks. Before and after treatment,the body weight of rats was recorded,the pathological changes of ovarian tissue in rats were observed,the serum levels of testosterone( T) and fasting insulin( FINS)were detected and the insulin sensitivity index( ISI) was calculated. Results After HE staining,multiple follicles at different developmental stages were observed in rats of the control group,as well as the multilayered granular cells. The multiple follicular cystic dilatation was observed in the ovarian tissue of rats in the model group,with less granular layer and thickening albuginea. In the electroacupuncture group,the cystic follicles were decreased and the granular layer was increased in the ovarian tissue of rats. Compared with the control group,the body weight,levels of T and FINS were significantly increased in the model group,and the ISI was significantly decreased( P<0.05,P < 0.01). Compared with the model group,the body weight and T level were significantly decreased in the electroacupuncture group( P<0.05),and the FINS level was decreased and the ISI was increased,with no significant differences. Conclusion Electroacupuncture can decrease the body weight and serum T level in PCOS-IR rats,and improve the insulin resistance to some extent.
