四川桤木上胚轴的离体培养与植株再生 被引量:3

摘要 通过直接诱导四川桤木(AlnuscremastogyneBurkill)幼芽上胚轴分化不定芽的途径获得了再生植株,比较了不同植物生长调节物质、糖类及其浓度在离体培养中的效应。认为四川桤木不定芽的分化需经过附加6-BA的WPM培养基的预培养和移植培养2个阶段;经45d的预培养和20d的移植培养,每个外殖体平均可以获得3 4~9 8个不定芽。培养基中糖的种类及浓度对不定芽的诱导有显著的影响,认为3%的蔗糖和葡萄糖适宜于增殖培养基的碳源;生根受生根培养基、增殖培养基中糖的种类和浓度的影响,以3%的葡萄糖为碳源适宜于不定芽及其根系的分化和生长,生根率可达到93 3%~100%;在蛭石基质中炼苗成活率可达100%。 The regeneration plants were obtained through the way of inducing directly the epicotyl of Alnus cremastogyne juvenile bud to differentiating into adventitious buds,this paper compared the effects of different plant growth conditioning substances,saccharides and saccharide concentrations in the isolated culture.It believeed that the A.cremastogyne adventitious bud differentiation should finished through 2 periods of the culture beforehand and transplant culture of WPM culture medium of additional 6-BA;34~98 adventitious buds were obtained avergely per explant through 45 days long culture beforhand and 20 days long transplant culture. 
