电针介入时机对腰多裂肌损伤模型大鼠Foxo1、Myostatin、Myod蛋白表达的影响 被引量:8

Effects of Electroacupuncture Intervention Time on the Expression of Foxo1,Myostatin and Myod in Rats with Multifidus Muscle Injury

摘要 目的:观察不同电针介入时机对腰多裂肌损伤大鼠多裂肌叉头蛋白(Foxo1)、肌肉生长抑制素(Myostatin)、成肌分化因子(Myod)蛋白表达的影响。方法:雄性SD大鼠随机分为空白组、模型组、即刻电针组、24 h电针组、48 h电针组,每组8只。以多裂肌肌注0. 5%布比卡因复制腰多裂肌损伤模型。各电针组分别在造模后即刻、24 h、48 h开始电针双侧"委中""肾俞",连续干预7 d。以Western blot检测各组多裂肌Foxo1、Myostatin、Myod蛋白表达水平。结果:与空白组比较,模型组Foxo1、Myod蛋白表达水平显著升高(P <0. 01);与模型组比较,即刻电针组、48 h电针组Foxo1、Myostatin蛋白表达水平降低(P <0. 01,P <0. 05),24 h电针组Foxo1、Myostatin蛋白表达水平显著降低(P <0. 01),Myod蛋白表达水平显著升高(P <0. 01);与24 h电针组比较,即刻电针组Myostatin蛋白表达水平升高(P <0. 05)、Myod蛋白表达水平显著降低(P <0. 01),48 h电针组Foxo1蛋白表达水平显著升高(P <0. 01)、Myod蛋白表达水平显著降低(P <0. 01)。结论:电针促进多裂肌修复的最佳介入时机可能为损伤后24 h。 Objective: To observe the effects of different electroacupuncture intervention time on the expression of multifidus muscle fork protein( Foxo1),Myostatin and myoblast differentiation factor( Myod) in rats with lumbar multifidus muscle injury. Methods: Male Sprague-Dawley rats were randomly divided into the blank group( BG),model group( MG),immediate/24 h/48 h electroacupuncture group( 0 h G/24 h G/48 h G),with 8 rats in each group. The lumbar multifidus muscle injury model was replicated with 0. 5% bupivacaine in the multifidus muscle. Each electroacupuncture group started the electroacupuncture on both sides of the Weizhong( BL40) and Shenshu( BL23) at 0 h/24 h/48 h after modeling,and continuous intervention for 7 days. Western blot was used to detect the expression levels of Foxo1,Myostatin and Myod proteins in multifidus muscles of each group. Results: Compared with the BG,the expression of Foxo1 and Myod proteins in the MG significantly increased( P < 0. 01);Compared with the MG,the expression of Foxo1 and Myostatin in the 0 h G and 48 h G were decreased( P < 0. 01,P < 0. 05). The expression of Foxo1 and Myostatin in the 24 h G significantly decreased( P < 0. 01),and Myod protein expression was significantly increased( P <0. 01). Compared with the 24 h G,the expression of Myostatin protein in 0 h G increased( P < 0. 05),and the expression of Myod protein in 0 h G significantly decreased( P < 0. 01);the expression of Foxo1 protein in 48 h G was significantly increased( P <0. 01),and the expression of Myod protein was significantly decreased( P < 0. 01). Conclusion: The best intervention time is 24 h after injury,wich can effectively promote multifidus muscle repairing.
