六白白疕巴布剂血竭素含量测定及样品提取方法的研究

摘要 目的研究六白白疕巴布剂中血竭素含量测定及供试品溶液提取方法。方法以血竭素提取转溶率为指标,通过单因素考察,并在单因素考察的基础上,再对提取次数、提取时间和溶剂用量等三个因素进行正交设计实验,确定该巴布剂样品血竭素最佳提取转溶方法。结果单因素考察结果表明,膏体取样量约2 g时,20 ml氨试液用量可以基本解离血竭素甘草酸盐;密闭振摇提取效果优于回流提取和超声振动提取;振摇提取3 mim和4 min比较差异无统计学意义(P>0.05);三氯甲烷单次提取用量15 ml和20 ml比较差异无统计学意义(P>0.05)。正交实验结果表明,取约2 g膏体量时,每次用10 ml三氯甲烷密塞振摇提取2次,每次提取时间3 min,可以基本溶解转溶该巴布剂中血竭素成分。结论该提取方法可以作为该巴布剂样品血竭素提取转溶方法,提取后残余膏体已基本检测不出血竭素成分。 Objective To study the extraction method for dracorhodin determination in the sample solution of liu bai bai bi cataplasm. Methods The dracorhodin extraction and solubilization rate was taken as the index. Through single factor investigation,and on the basis of single factor investigation,the orthogonal design test was conducted on the three factors,named extraction frequency,extraction time and solvent dosage. And the best extraction solution method was determined from cataplasm sample of dracorhodoin. Results The single factor investigation results showed: when the paste sample was about 2 g,dracorhodin glycyrrhetate could be dissociated from 20 ml ammonia test solution. The result of airtight shake extraction was better than that of reflux extraction and supersonic vibration extraction. The results were not different significantly between 3 min shake extraction and 4 min shake extraction( P > 0. 05). The results were not different significantly in the chloroform extraction between 15 ml and 20 ml( P > 0. 05). The orthogonal test results showed:for about 2 g paste,the two- time airtight shake extraction with 10 ml chloroform,3 min for each time,dracorhodin component was basically dissolved from the cataplasm. Conclusion This method can be used for the extraction and solubilization of dracorhodin from this cataplasm and the dracorhodin component is not basically detected from the residue.
