葛根汤对去势MCAO雌性小鼠模型的保护作用研究

Protective Effect of Gegen Decoction in Ovariectomized MCAO Female Mouse Models

目的观察葛根汤对去势雌性小鼠大脑中动脉阻塞模型(MCAO)脑损伤的保护作用,为葛根汤治疗绝经后妇女脑供血不足的临床应用提供实验依据。方法采用摘除双侧卵巢(OVX)复合大脑中动脉阻塞法(MCAO)复制去势雌性小鼠脑缺血模型,实验随机分为假手术组,模型组,葛根汤低、中、高剂量组(i.g.3.5 g/kg,7 g/kg,14 g/kg)和阳性对照组(苯甲酸雌二醇,i.p.10 mg/kg),通过Longa的5分制法评分标准和TTC染色法分别进行神经学评分和计算脑梗死面积,同时测定脑含水量。结果葛根汤不同剂量均能够有效改善模型组小鼠的行为学障碍,降低神经学评分,各给药组与模型组比较,差异均有统计学意义(P<0.05);葛根汤中、高剂量组小鼠脑梗死面积减小,与模型组比较,差异均有统计学意义(P≤0.05);葛根汤对脑含水量的影响并不明显。结论葛根汤对去势雌性小鼠缺血性脑损伤具有保护作用,为其治疗绝经后妇女脑供血不足的临床应用提供了参考依据。

Objective To observe the protective effect of gegen decoction of MCAO brain damage in ovariectomized female mouse models so as to provide the experimental evidence for gegen decoction in treatment of brain blood supply insufficiency in postmenopausal women. Methods The OVX and MCAO were used to duplicate brain ischemia models of oxariectomized female mice. The pseudo- operation group,model group,low- dose,middle- dose and high- dose groups( i. g. 3. 5 g / kg,7 g / kg,14 g / kg,gegeng decoction)and positive control group( estradiol benzoate,i. p. 10 mg / kg) were randomized. Longa 5- credit system scale and TTC staining were used for neurological evaluation and calculation of brain infarction area,as well as the determination of brain water content. Results Gegen decoction of different doses effectively relieved the behavior disturbance and reduced neurological score in the mice of the model group,indicating the significant differences as compared with that in each medication group( P < 0. 05). In the middle- dose and high- dose group,the brain infarction area was reduced,indicating the significant difference as compared with the model group( P≤0. 05). Gegen decoction did not impact apparently brain water content. Conclusion Gegen decoction has the protective effect on brain ischemic damage in ovariectomized female mice,which provides the experimental evidence for gegen decoction in treatment of brain blood supply insufficiency in postmenopausal women.
