新辅助化疗对骨肉瘤患者外周血淋巴细胞HSP72表达的影响及意义

Effect of Neoadjuvant Chemotherapy on Expression of HSP72 Protein in Lymphocyte of Osteosarcoma Patients

目的观察新辅助化疗对骨肉瘤患者外周血淋巴细胞热休克蛋白72(HSP72)表达的变化及临床意义。方法收集9例骨肉瘤患者化疗前后外周血为实验组,同时以11例健康献血者为对照组。应用RT-PCR和Western blot技术检测骨肉瘤患者化疗前后外周血淋巴细胞HSP72 mRNA和蛋白的表达,并同时检测对照组外周血淋巴细胞HSP72 mRNA和蛋白的表达。结果实验组外周血淋巴细胞HSP72 mRNA和蛋白的表达明显高于对照组(P<0.01)。实验组化疗前HSP72 mRNA和蛋白的表达显著高于化疗后(P<0.05)。结论 HSP72在骨肉瘤患者化疗中的变化可作为评价骨肉瘤对化疗药物敏感性的一个指标。

Objective To observe the effect of neoadjuvant chemotherapy on expression of heat shock protein 72( HSP72) protein in peripheral blood lymphocytes of osteosarcoma patients. Methods Nine patients with osteosarcoma were included in the experimental group,and 11 cases of healthy blood donors were in the control group. RT- PCR and Western blot methods were used to detecte the HSP72 mRNA and protein expression on peripheral blood lymphocytes in patients with osteosarcoma before and after chemotherapy,and expression of HSP of peripheral blood lymphocytes in control group were also detected. Results HSP72 mRNA and protein expression on experimental group were significantly higher than those of the control group( P < 0. 01). Expression of HSP72 mRNA and protein in pre- chemotherapy were significantly higher than post- chemotherapy( P < 0. 05). Conclusion HSP72 expressed in patients with osteosarcoma may evaluate osteosarcoma sensitivity to chemotherapy drugs,which can guide the application of clinical drug.
