摘要
目的观察活血四妙汤对高尿酸血症大鼠血尿酸(SUA)及黄嘌呤氧化酶(XOD)的影响,探讨其防治高尿酸血症的机制。方法 60只SD雄性大鼠随机分为正常组、模型组、别嘌呤醇组、活血四妙汤小剂量组、活血四妙汤中剂量组、活血四妙汤大剂量组,除正常组外,其余5组均采用灌胃腺嘌呤和盐酸乙胺丁醇复制高尿酸血症模型,于造模当日起,别嘌呤醇组予别嘌呤醇混悬液40 mg/kg,活血四妙汤小剂量组予活血四妙汤7 g/kg,中剂量组予10.5 g/kg,大剂量组予14 g/kg,正常组及模型组每日灌胃0.9%氯化钠注射液3 m L/只,于造模及给药14 d后采血并取材。应用比色法检测血尿酸值,应用ELISA检测XOD值。结果模型组与正常组相比血尿酸值、XOD显著增高(P<0.05)。与模型组比较,活血四妙汤大剂量组、中剂量组血尿酸值、XOD显著下降(P<0.05),活血四妙汤小剂量组与模型组相比血尿酸值、XOD差异无统计学意义(P>0.05)。活血四妙汤小剂量、中剂量、大剂量组的血肌酐值(Scr)、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)与正常组相比差异无统计学意义(P>0.05)。结论活血四妙汤可以降低高尿酸血症大鼠血尿酸,并存在剂量依赖性,其降低尿酸机制与调节XOD的血液浓度成正相关,且活血四妙汤经胃肠途径给药无肝肾毒性。
Objective To investigate the effects of Huoxue Simiao decoction on hyperuricemia rat serum uric acid and Xanthine oxidase,and to explore the mechanisms of prevention and treatment of hyperuricemia. Methods Adenine and ethambutol were fed to establish hyperuricemia early renal damage models.Sixty SD male rats were randomly divided into normal group,model group,allopurinol group,Huoxue Simiao Formula low- dose group,Huoxue Simiao Formula medium dose group,Huoxue Simiao Formula high- dose group. Except the normal group,the other five groups all established the hyperuricemia early renal damage models,from the same day,the allopurinol group fed rats with 40 mg / kg of allopurinol suspension,the Huoxue Simiao Formula low- dose group fed 7 g / kg of Huoxue Simiao Formula,the medium dose group fed 10. 5 g /kg,high- dose group fed 14 g / kg,and normal group and model group fed rats with 0. 9% sodium chloride injection 3 ml. On the modeling day and the 14 thday,blood was drawn. Colorimetry Colourimetry was used to detect uric acid and ELISA was applied to measure XOD level. Results XOD and uric acid level of model group were significantly higher compared with normal( P < 0. 05); compared with the model group,uric acid and XOD level in Huoxue Simiao Formula high dosage group and medium dosage group decreased significantly( P < 0. 05); compared with the model group,Huoxue Simiao Formula low dosage group showed no significant differences in terms of uric acid and XOD level( P > 0. 05); The level of serum creatinine,AST,ALT among Huoxue Simiao Formula high dosage group,medium dosage group and low dosage group showed no significant differences compared to the normal group( P > 0. 05). Conclusion Huoxue Simiao Formula can reduce uric acid on hyperuricemia rat and depend on dosage,and blood levels of uric acid lowering mechanism is in positive correlation with regulation XOD. Huoxue Simiao Formula administration shows no toxicity to the liver and kidney.
