摘要
目的观察糖脂平对高脂饮食诱导的胰岛素抵抗(IR)大鼠血脂及脂肪因子的影响。方法将80只雄性SD大鼠随机分为正常组(20只)和造模组(60只),造模组用高脂高糖饲料喂养8周造模,其中54只造模成功的大鼠分为模型组、中药组、西药组,每组18只。中药组和西药组分别给予糖脂平和二甲双胍灌胃,模型组和正常组灌服等量的生理盐水。给药8周后,用ELISA测定各组大鼠的血清脂联素、瘦素、抵抗素水平,同时检测血脂和游离脂肪酸(FFA)水平。结果与正常组相比,模型组大鼠肥胖明显,总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、FFA均明显增高,高密度脂蛋白胆固醇(HDL-C)显著降低,表明模型组大鼠发生了脂代谢紊乱。同时脂肪因子、瘦素和抵抗素水平显著升高,脂联素水平显著降低。与模型组比较,糖脂平可以降低TG、LDL-C和FFA、瘦素、抵抗素水平,提高脂联素水平,对TC和HDL-C无明显影响。结论糖脂平可改善IR大鼠血脂紊乱,调节脂肪因子水平。
Objective To observe the impact of tangzhiping on blood lipid and adipokine in the rats of insulin resistance(IR)induced by high fat diet. Methods Eighty male SD rats were randomized into a normal group(20 rats)and a model group(60 rats). In the model group,the high - fat and high - sugar diet was used to prepare the models for 8 weeks. Totally,54 of 60 rats were modeled successfully and randomized into a model group,a Chinese medicine group and a western medicine group,18 rats in each one. In the Chi-nese medicine group and western medicine group,tangzhiping and metformin for lavage respectively and in the model group and normal group,the physical saline of the same dose was used. In 8 weeks of medication, ELISA was used to determine the levels of serum adiponectin,leptin and resistin,as well as the levels of blood lipid and free fat acid(FFA). Results Compared with the normal group,the rats were obese apparent-ly in the model group,TC,TG,LDL - C and FFA were increased apparently and HDL - C was reduced signif-icantly,which indicated that the rats in the model group had lipid metabolic disorder. At the same time,the levels of serum leptin and resistin were increased apparently. Adiponectin was reduled significantly in the model group. Compared with the model group,tangzhiping reduced TG,LDL - C,FFA,leptin and resistin lev-els,increased adiponectin level and had no apparent impact on TC and HDL - C. Conclusion Tangzhiping improves in the blood lipid disorders and regulates the adipokine levels in IR rats.
